褪黑素对带状疱疹后神经痛大鼠血清CD4^+ CD25^+调节性T细胞表达的影响 被引量:4

The Influence of Melatonin on CD4^+ CD25^+ Regulatory T Cell Expression in Serum of Postherpetic Neuralgia Rats

摘要 目的:观察褪黑素对带状疱疹后遗神经痛(PHN)大鼠CD4^+ CD25^+调节性T细胞表达的影响。方法:雄性SD大鼠60只,随机分为空白对照组(不做任何处理)、假手术组(仅暴露松果体)、PHN模型组(松果体切除后接种带状疱疹病毒)及治疗组(与PHN模型组一样造模,同时腹腔注射外源褪黑素);接种病毒30 d时进行机械痛觉评分,同时取大鼠外周血应用流式细胞术检测CD4^+ CD25^+调节性T细胞表达,应用RT-PCR技术检测Foxp3+基因表达,采用ELISA技术检测外周血TGF-β表达。结果:接种病毒30 d时,空白对照组与假手术组比较各项指标差异均无统计学意义(P>0.05);模型组和治疗组大鼠痛觉评分、外周血CD4^+ CD25^+调节性T细胞、Foxp3+基因和TGF-β表达均显著高于正常对照组(P<0.05);模型组各项指标均显著高于治疗组(P<0.05)。结论 :腹腔注射褪黑素可减轻带状疱疹病毒感染大鼠的PHN,其机制可能与下调CD4^+ CD25^+调节性T细胞表达改善机体免疫功能有关。 Objective: To observe the influence of melatonin on β regulatory T cell ex- pression in postherpetie neuralgia (PHN) rats. Method : Sixty male SD rats were randomly divided in- to 4 groups: blank control group (group B), sham-operation group (group S), PHN model group (group P), and treatment group (group T). Rats in group A were not dealt with specially. Conarium of rats in group B were exposed. Conarium of rats in group P were removed and the rats were inocula- ted with herpes zoster virus. Rats in group T were inject with exogenous menatonin intraperitoneally af- ter received operations of group P. In 30 day after virus inoculation, mechanical pain was graded, ex- pression of CD4+CD25+ regulatory T cells in rat peripheral blood (PB) was tested with flow cytome- try, Foxp3 + gene expression detected with RT-PCR technique, and PB TGF-13 expression determined using ELLISA technology. Result: All of the indicators were not significantly different between groups B and S(P 〉 0.05). The pain scores, expressions of PB CD4+CD25+ regulatory T cells, Foxp3 + gene, and TGF-β were significantly higher in groups P and T than in group B ( P 〈 0.05 ) , and were markedly higher in group P than in group T. Conclusion: Peritoneal cavity injection of melatonincould relieve PHN in herpes zoster virus infected rats. The mechanism might relate with the decrease of expression of CD4+CD25+ regulatory T cells and the improvement of immunity.
