摘要
目的:探讨白藜芦醇对基质金属蛋白酶-2活性及耐脱矿力的影响,以期为白藜芦醇的临床应用提供实验依据。方法:设置实验组白藜芦醇的浓度梯度:12.5μmol/L、25μmol/L、50μmol/L、100μmol/L、200μmol/L、400μmol/L,阳性对照组0.2%氯己定,阴性对照组去离子水。每组取10μL与提取出来的牙源性mmp-2 30μL混合孵育30min,用体液基质金属蛋白酶-2活性比色法定量检测各组混合20 min后的酶活性,每组重复5次。牙本质片经pH循环后,用激光共聚焦电子显微镜3D图像观察牙本质脱矿深度。结果:实验组白藜芦醇浓度在50μmol/L以上时有明显抑制作用,且在100μmol/L、200μmol/L、400μmol/L时的酶活性抑制率与阳性对照组间无显著性差异。激光共聚焦电子显微镜3D图像显示100μmol/L Res的染色深度明显低于阴性对照组,统计学分析结果显示100μmol/L Res的平均脱矿深度显著低于阴性对照组(P<0.05),与阳性对照组间并无显著性差异。结论:白藜芦醇可抑制基质金属蛋白酶-2活性及牙本质脱矿。
Objective:To discuss the effects of resveratrol on matrix metalloproteinase-2(mmp-2)activity and demineralization resistance and to provide experimental evidence for the clinical application of resveratrol.Methods:The concentration gradients of resveratrol were 12.5μmol/L,25μmol/L,50μmol/L,100μmol/L,200μmol/L,and 400μmol/L in the experimental group.0.2% chlorhexidine was selected as the positive control group and deionized water as the negative control group.10μL of each group was mixed with 30μL extracted odontogenic mmp-2and incubated for 30 min.Metalloproteinase-2activity was quantitatively detected with body fluid matrix metalloproteinase-2activity assay after 20 min,and each group was repeated five times.After going through the pH cycle,electron microscopy and laser scanning confocal 3Dimage were used to observe dentin demineralization depth.Results:Resveratrol with the concentration above 50μmol/L showed significant inhibitory effect.And metalloproteinase-2activity inhibition rate in groups with the concentration of 100μmol/L,200μmol/L,and 400μmol/L showed no significant difference compared to the positive group.Confocal electron microscopy 3Dimage displayed that Res dyeing depth in the 100μmol/L group was significantly lower than that in the negative control group.And statistical analysis showed the same results(P〈0.05).But there was no significant difference between 100μmol/L group and the positive control group.Conclusion:Resveratrol can inhibit the activity of matrix metalloproteinase-2and dentin demineralization.
