毛细管电泳与高效液相色谱法在HbQ-Thailand检测中的比较研究 被引量:3

摘要 目的了解毛细管电泳(CE)与高效液相色谱法(HPLC)在Hb Q-Thailand检测中的差异。方法收集10例CE筛查出的可疑Hb Q-Thailand,经地贫基因分析和DNA测序确诊后,再用HPLC检测比对。结果 10例样本中CE均检出Hb Q-Thailand和Hb Q-Thailand A2(Hb QA2),而HPLC仅检出10例Hb QThailand和5例Hb QA2,而且HPLC错误检出Hb Q-H病1例存在Hb A2。CE和HPLC检出Hb Q-Thailand、Hb A2、Hb QA2的含量分别为(34.41±21.10)%、(1.68±0.57)%、(0.77±0.10)%和(31.72±21.46)%、(2.43±0.22)%、(0.24±0.24)%,两组Hb Q-Thailand差异无统计学意义(P>0.05),Hb A2和Hb QA2的差异有统计学意义(P<0.01)。结论 CE在检测Hb Q-Thailand中优于HPLC。 Objective To study the difference in detection of Hb Q-Thailand between capillary electrophoresis( CE) and high performance liquid chromatography( HPLC). Methods Ten samples of suspicious Hb Q-Thailand by CE were confirmed with thalassemia gene analysis and DNA sequencing and their results were compared with those detected by HPLC. Results Ten samples with Hb Q-Thailand and Hb Q-Thailand A2( Hb QA2) were all detected by CE,but Hb Q-Thailand and Hb QA2 were detected respectively in 10 samples and 5 samples by HPLC. Moreover,Hb A2 was wrongly detected in Hb Q-H disease by HPLC. The values of Hb Q-Thailand,Hb A2 and Hb QA2 detected by CE and HPLC were( 34. 41 ± 21. 10) %,( 1. 68 ± 0. 57) %,( 0. 77 ± 0. 10) % and( 31. 72 ± 21. 46) %,( 2. 43 ± 0. 22) %,( 0. 24 ± 0. 24) % respectively. There were significant differences in the detection of Hb A2 and Hb QA2 between the different methods of CE and HPLC( P〈0. 01),but there was no significant difference in the detection of Hb Q-Thailand between the two methods( P〉0. 05). Conclusion CE is better in detection of Hb Q-Thailand than HPLC.
