妊娠期高血压疾病AGEs、RAGE、TGF-β1的表达 被引量:26

Expression of AGEs、RAGE、TGF-β1 in hypertensive disorders complicating pregnancy

摘要 目的:检测孕中期和孕晚期外周血和胎盘晚期糖基化终末产物(AGEs)及其受体RAGE、转化生长因子-β1(TGF-β1)的表达,探讨各指标与妊娠期高血压疾病的关系。方法:2014年8月至2015年10月在南通大学附属医院产科门诊产前检查做唐氏筛查的孕妇血清样本900例,追踪孕妇的妊娠结局,获取妊娠期高血压9例,子痫前期(PE)组6例。检测孕妇孕中期和孕晚期血清和胎盘中AGEs、RAGE、TGF-β1表达。结果:孕中期,PE组血清AGEs、RAGE、TGF-β1水平比对照组明显升高(P<0.05),妊娠期高血压组TGF-β1水平也较对照组明显升高(P<0.05)。孕晚期,妊娠期高血压组和PE组AGEs、TGF-β1水平比对照组均明显增高,PE组TGF-β1表达水平比妊娠期高血压组显著升高(P<0.05)。PE组RAGE表达水平比对照组和妊娠期高血压组均显著升高(P<0.05)。PE胎盘AGEs、RAGE、TGF-β1表达阳性染色细胞比对照组和妊娠期高血压组染色明显增强,数量明显增多。结论:AGEs-RAGE系统的激活可能和PE的发生密切相关,TGF-β1参与了这个病理过程。 Objective:To investigate the relationship between expression of advanced glycation end product( AGEs),its receptor RAGE,transforming growth factor-β1( TGF-β1)and hypertensive disorders complicating pregnancy.Methods:Samples from 900 pregnant women who presented to the Affiliated Hospital of Nangtong University for prenatal care from Aug.2014 to Oct.2015 were included in this prospective study,of which 15 were subsequently developed hypertensive disorders complicating pregnancy as case group( preeclampsia 6 and gestational hypertension 9) and 15 were normal pregnant women as control.The matemal serum was conducted in their second( between 16 th to 20 th week of pregnancy) and third( between34 th to 36 th week of pregnancy) trimester of pregnancy.Expression of serum AGEs,RAGE,TGF-β1 were detected by Elisa.Expression of AGEs,RAGE,TGF-β1 in the placentas were detected by immunohistochemical staining.Result:In the second trimester of pregnancy,the expression levels of AGEs,RAGE,TGF-β1 in the peripheral blood of pregnant women with preeclampsia were significantly higher than those in the control group( P 0.05).The expression level of TGF-β1 in gestational hypertension was significantly higher than that in the control group( P0.05).In the third trimester of pregnancy,the expression levels of AGEs and TGF-β1in the peripheral blood of pregnant women with preeclampsia and gestational hypertension were both significantly higher than those in the control group( P0.05),while the expression level of Rage in preeclampsia was significantly higher than those in gestational hypertension and the normal control group.( P0.05).Positive expressions of AGEs,RAGE,TGF-β1 were detected in the placenta of preeclampsia.Positive correlations were observed among AGEs,RAGE,TGF-β1.Conclusions:Expression of AGEs,RAGE,TGF-β1 are associated with preeclampsia.
