摘要
将一株从大庆油田含油土壤中分离筛选得到的专一性脱硫力最高的菌株HNHHJ-1作为培养菌株,通过对DBT转化为2-HBP的脱硫效率来了解初始p H值、氮源、碳源、培养温度和摇瓶转速条件对脱硫力的影响,并优化培养条件。确定的最佳培养条件为:在30℃温度下培养,每250m L三角瓶装100mL培养基,接种量为1%(V/V),转速为180r·min^(-1)。在上述培养条件下培养,2-HBP生成量为22.806mg·L^(-1)。
A high specificity desulfurization strain HNHHJ-1 which was isolated from the Daqing Oilfield oily soil had been studied. By comparing the efficiency of conversion dibenzothiophene (DBT) into 2-hydroxybiphenyl (2-HBP), the influence of initial pH value, nitrogen source, carbon source, culture temperature and shake calabash speed on desulfurization conditions were investigated. The optimal culture conditions of HNHHJ-1 were finally de- termined. The results showed that the best culture conditions as follows: 100 mL nutrient solution containing in ev- ery 250 mL conical flask, the inoculation of biomass was 1%, the speed was 180r·min-1, and the temperature was 30℃. In above optimum conditions, the yield of 2-HBP arrived at 22.806 mg·L-1.
