犬小肠缺血再灌注后血中NO和SOD与小肠粘膜凋亡相关基因表达的改变 被引量:4

摘要 研究犬小肠缺血再灌注后氧自由基和c fos、增殖性细胞核抗原 (PCNA)及Bax的表达改变及其相关性。于阻断小肠动脉前及再灌注 0、30、及 6 0min采集伴行静脉血液标本及小肠组织进行各项指标检测。结果显示 ,再灌注 0min ,NO和SOD的血中浓度明显下降 ,c fos、PCNA和Bax在小肠粘膜上皮及小肠腺的表达明显增强。再灌注 30min ,NO和SOD浓度降至最低、c fos、PCNA和Bax的表达达最高峰。至再灌注 6 0min ,NO和SOD有所升高 ,而三种基因的表达则显著减少。NO和SOD的改变与三种基因表达的相关性分析显示出他们之间密切的相关关系。犬小肠缺血再灌注可造成氧自由基的增多和c fos、PCNA和Bax的表达增加 ,可能与损伤后的修复重建等有关 。 The changes and correlation between oxygen free radical and c fos, PCNA and Bax in intestinal ischemia reperfusion of dogs would be studied. We clamped the intestinal artery, collected the blood sample from accompanying intestinal vein before clamping the artery and reperfusion for 0,30,60 min, and took intestinal tissue for various items inspect. The results showed that after reperfusion 0 min, the concentration of NO and SOD decreased, and the expression of c fos, PCNA and Bax increased obviously. After reperfusion 30 min, the concentration of NO and SOD was at the lowest level, and the expression of c fos, PCNA and Bax increased to a peak. But after reperfusion 60 min the concentration of NO and SOD increased obviously and expression of c fos, PCNA and Bax all decreased significantly. Correlation analysis of the changes of oxygen free radical and the expression of c fos, and Bax showed their close correlativity. Ischemia reperfusion of small intestine of dog might increase the concentration of oxygen free radical and the expression of c fos, PCNA and Bax which probably play an important role in recovery and reconstruction of injured intestine,and concentration of oxygen free radical had a close relationship with expression of c fos, PCNA and Bax.
