Analysis of VP1 gene of enterovirus 71 isolated in Kunming and Qujing city from 2012 to 2013

摘要 目的分析2012-2013年间,云南省昆明、曲靖两地区手足口病(hand,foot and mouth disease,HFMD)患者中肠道病毒71型(enterovirus 71,EV-A71)的基因特征。方法对收集的病例粪便样本进行病毒分离培养和real-time RT-PCR鉴定,并对鉴定为EV-A71型的毒株进行VP1区的核酸序列测定,构建遗传进化树进行基因型别分析。结果两地两年间共收集了538份样本,其中EV-A71型分离株105份,105份EV-A71型分离株均属于C4a亚型,105株EV-A71型分离株在遗传进化树明显聚集为三簇,提示在两地存在多种病毒传播链的可能。结论 2012-2013年昆明、曲靖两地由EV-A71引起的手足口病均属C4a亚型,但其存在多条传播链的流行趋势。 Objective To analyze the genetic characteristics of enterovirus 71(EV-A71)strains isolated from cases of hand,foot and mouth diseases(HFMD)in Kunming and Qujing city from 2012 to 2013in China.Methods Enterovirus isolation was performed in human malignant rhabdomyosarcoma cells(RD)and Hep-2cell lines from the collected stool specimens.Type EV-A71 strains were identified by real time RT-PCR from the positive cultures. Results A total of 105EV-A71 strains were isolated and identified from 538 specimens in this study.The complete VP1 sequences of 105EV-A71 strains were determined.These EV-A71 strains were grouped to C4 asubtype,which were co-evolved with isolates from other areas in mainland of China.Phylogenetic tree analysis revealed that 105EV-A71 strains were clustered into 3branches. Conclusions HFMD in Kunming and Qujing city from 2012 to 2013in China was caused by EV-A71C4 asubtype.
