摘要
目的探讨健脾补肾法对骨髓抑制小鼠造血功能的影响。方法取BALB/c小鼠60只,按照体重将60只雄性小鼠随机分为正常对照组、模型组、阳性对照组、健脾补肾法组,每组15只。模型组、阳性对照组、健脾补肾法组小鼠按照0.1m L/10 g体重腹腔注射8 mg/m L环磷酰胺溶液(剂量为80 mg/kg),正常对照组小鼠按照0.1 m L/10 g体重腹腔注射0.9%氯化钠注射液,每天1次,连续3天。确定小鼠造模成功后,健脾补肾法组小鼠每日上下午分别按照0.2 m L/10 g体重灌胃给予2g/m L健脾补肾方及龟鹿二仙丹,阳性对照组小鼠每日按照0.1 m L/10g体重腹腔注射12.5μg/m L rh G-CSF,每天1次,连续给药7天。正常对照组和模型组均按照0.2 m L/10 g体重灌胃给予纯净水。末次给药后24 h,经小鼠眼球后静脉丛采血,用装有EDTA-Na抗凝剂的采血管收集,用全自动血细胞分析仪进行白细胞检测。颈椎脱臼处死各组小鼠,检测骨髓造血细胞集落形成和骨髓有核细胞的情况。结果健脾补肾法具有促使骨髓抑制小鼠骨髓细胞解除Go/G1期阻滞、促使Go/G1期细胞向S期细胞以及加速S期细胞向G2/M期细胞转化,从而促进骨髓细胞增殖的作用。结论健脾补肾法对化疗后所致骨髓抑制有显著的改善和恢复作用。
Objective To discuss the influence of Jianpi Bushen recipe on the blood-prodhcing function in mice of myelosuppression induced by cyclophosphamide. Methods Taking BALB/c mice 60 cases, according to the weight, 60 male mice were randomly divid-ed into normal control group, model group, positive control group and Jianpi Bushen recipe group, with 15 cases in each group. The model group, the positive control group and Jianpi Bushen recipe group were given intraperitoneal injection of 0.1 m L / 10 g weight 8mg/m L solution cyclophosphamide(dose of 80 mg/kg). The normal control group mice was given intraperitoneal injection of 0.1 m L/ 10 g weight 0.9% sodium chloride injection, 1 time a day for three days. After the success of the building, the mice in Jianpi Bushen recipe group were given daily 0.2 m L / 10 g, weight lavage for 2 g/m L Jianpi Bushen recipe gr and Guilu Erxian minipills in the morning and afternoon. The positive control group mice were given intraperitoneal injection of 0.1 m L / 10 g weight 12.5 mu g/m L rh G CSF, 1 time a day for seven days. The normal control group and model group were in accordance with 0.2 m L / 10 g weight to fill the stomach to pure water. The last 24 h after the treatment, venous plexus after eyeball in mice blood, usedh EDTA-Na anticoagulant blood vessels of the adopt of collection, white blood cell detection using automatic blood cell analyzer. Cervical dislocation exe-cution groups of mice, detection of bone marrow hematopoietic cell colony formation and bone marrow nucleated cells. Results Jianpi Bushen recipe method have prompted bone marrow inhibition of mouse bone marrow cells remove Go/ G1 phase retardation, prompted Go/ G1 phase to S phase cells, and accelerated the S phase cells to the G2/ M phase transformation, so as to promote the role of bone marrow cell proliferation. Conclusion Jianpi Bushen recipe can significantly improve and restore function of bone marrow suppression after chemotherapy.
