补肾壮骨方治疗大鼠骨质疏松症效果的实验研究 被引量:5

摘要 目的通过运用自拟补肾壮骨方治疗骨质疏松症,观察大鼠的体重、骨密度、骨生物力学等一般情况变化,并对血清OCN、PICP、25-(OH)D3,1,25-(OH)2D3等指标影响进行研究。方法采用SPF级雌性大鼠,80只,按体重进行分层随机分为正常组、模型组、补肾壮骨方治疗组、阿仑膦酸钠阳性药物观察组。采用摘除卵巢对非正常组大鼠进行骨质疏松症造模,正常组和模型组给予1 m L/100 g的生理盐水,日2次,灌胃;补肾壮骨方治疗组给予补肾壮骨中药汤剂(1 m L/100g,1 m L含1 g生药),日2次,灌胃;阿仑膦酸钠阳性药物观察组给予阿仑膦酸钠溶液(1 m L/100 g,1 m L含0.74 mg药粉),日2次,灌胃。连续灌胃3个月。观察大鼠股骨重量、骨密度变化、骨生物力学变化、血清OCN、PICP、25-(OH)D3、1,25-(OH)2D3等指标变化。结果大鼠股骨重量骨密度变化;骨生物力学变化;血清OCN、PICP、25-(OH)D3,1,25-(OH)2D3等指标;实验后,与正常组比较,模型组大鼠各项指标改变明显(P<0.05),差异具有统计学意义;补肾壮骨中药治疗组、阿仑膦酸钠阳性药物观察组与模型组大鼠比较,均改善明显(P<0.05),补肾壮骨方治疗组与阿仑膦酸钠观察组比较股骨重量(右侧)、骨生物力学、血清OCN、PICP、血清25-(OH)D3、1,25-(OH)2D3变化明显(P<0.05),具有统计学意义,骨密度变化不明显(P>0.05),差异无统计学意义。结论补肾壮骨中药治疗骨质疏松症模型大鼠效果明显,能够改善大鼠股骨重量、骨密度、骨生物力学、血清OCN、PICP、25-(OH)D3,1,25-(OH)2D3等,通过多靶点、多途径的促进骨细胞增殖,骨形成与重建,骨钙磷代谢。 Objective By using self-made Bushen Zhuanggu decoction in the treatment of osteoporosis, to observe generally changes of rats weight, bone mineral density, bone biomechanics, and to study the indexes of Serum Osteocalcin( OCN), PICP and 25-( OH) D31, 25-( OH)2D3etc. Methods 80 female rats of SPF, according to body weight, were randomized to normal group, model group, Bushen Zhuanggu decoction treatment group and alendronate sodium positive drug observation group. The introduction of o-variectomized rats in the non normal group of osteoporosis rats, normal group and model group were given physiological saline 1ml /100 g, 2 times a day, gavage. Bushen Zhuanggu decoction treatment group was given Bushen Zhuanggu Chinese medicine decoction(1ml / 100 g, 1ml contains 1g crude drug), 2 times a day, gavage. Alun phosphonic acid sodium positive drug observation group was treated with alendronate sodium solution(1ml / 100 g, 1ml contains 0.74 mg powder), 2 times a day, gastric lavage. The treatment continued irrigation for 3 months. The changes of the weight, bone mineral density, bone biomechanical changes, serum OCN, PICP,25-(OH) D3, 1, 25-(OH)2D3and other indexes in rats were observed. Results The changes of rat femur weight bone density, bone biomechanics, serum OCN, PICP, 25-(OH) D31, 25-(OH)2D3index after experiment were compared with the normal group, the indexes of model group rats changed significantly( P〈 0.05), and there was statistical significance. The Bushen Zhuanggu treatment group and the alendronate positive drug observation group was compared with the model group, and the improvement was significant( P〈 0.05).Bushen Zhuanggu decoction treatment group and alendronate observation group was compared the weight of femur( right), bone biomechanics, serum OCN, serum PICP, 25-( OH) D3, 1,25-( OH)2D3, changes were significant( P〈 0.05), and there was statistical significance, but bone density did not change significantly( P〉 0.05), there was no statistical significance.Conclusion The Bushen Zhuanggu Chinese medicine in the treatment of osteoporosis model rats has obvious effect, can improve the rat bone weight, bone density, bone biomechanics, Serum Osteocalcin( OCN), PICP and 25-( OH) D31, 25-( OH)2D3, through multiple targets and multiple ways to promote the proliferation of bone cells, bone formation and reconstruction, bone calcium and phosphorus metabolism.
