高效液相色谱法测定固体饮料中γ-氨基丁酸含量 被引量:2

摘要 建立了一种快速准确地测定固体饮料中γ-氨基丁酸含量的高效液相色谱分析方法:采用丹磺酰氯(Dansyl-Cl)为柱前衍生化试剂,色谱柱为Agilent Eclipse XDB-C18,流动相为25 mmol/L乙酸钠(p H=6.0)和乙腈的混合溶液(体积比为68∶32),流速为1.0 m L/min,柱温30℃,等度洗脱,紫外检测波长216 nm。结果表明,该方法的线性范围为0.002 5 mg/m L^0.1 mg/m L,R2=0.999 5,线性关系良好,检出限2.514 ng,平行试验6次,γ-氨基丁酸质量浓度的相对标准偏差(RSD)为0.89%,3个不同浓度的加标回收率在96.00%~102.39%之间,回收率相对标准偏差(RSD)在0.80%~2.87%之间。与传统的试验方法相比,该方法具有基线稳定,灵敏度高,线性范围广,回收率高及重现性好等特点。 A fast and accurate high performance chromatography method was developed for the determination ofγ-aminobutyric acid(GABA) in the solid beverages, by precolumn derivatization with dansyl chloride(Dansyl-Cl). The samples were separated on a Agilent Eclipse XDB-C18 at 30 ℃ using 25 mmol/L sodium acetate : acetonitrile(p H=6.0)=68:32 as mobile phase at a flow rate of 1.0 m L/min, isocratic elution, inspected by ultraviolet detector at 216 nm. The results showed that the γ-aminobutyric acid(GABA)has a good linear relationship at 0.002 5 mg/m L^0.1 mg/m L, the peak area with correlation coefficient at 0.999 5, and the detection limit was 2.514 ng. The relative standard deviation(RSD) of the mass concentration of six times was 0.89% for γ-aminobutyric acid(GABA). The three different concentrations of recovery was 96.00%~102.39%, and the relative standard deviation(RSD) was 0.80%~2.87%. Compared with the traditional experimental methods, utilizing isocratic elution, optimizing the detection wavelength and derivatization regent, the method has a stable baseline,high sensitivity,wider linear range,higher recovery and reproducible.
