右归丸对肾阳虚大鼠下丘脑-垂体-甲状腺轴的动态影响 被引量:9

摘要 目的动态研究右归丸对肾阳虚大鼠下丘脑-垂体-甲状腺轴的影响。方法 90只SD大鼠随机分成3组:正常组、模型组和右归丸组。采用氢化可的松构建肾阳虚模型,造模第8天开始,药物组用右归丸汤剂灌胃,正常组和模型组用生理盐水灌胃。分别在第15、23、30天处死动物,检测血清中促甲状腺素(T_SH)、三碘甲腺原氨酸(T_3)和四碘甲腺原氨酸(T_4)的含量,检测下丘脑匀浆中促甲状腺激素释放激素(T_RH)的含量,并对垂体、甲状腺质量进行称重和组织切片HE染色观察。结果与正常组比较,肾阳虚模型组大鼠各期T_RH、T_SH、T_3、T_4的含量明显降低(P<0.01);与模型组比较,右归丸组各期下丘脑-垂体-甲状腺轴激素水平均有升高,其中垂体-甲状腺轴激素水平升高明显(P<0.01),右归丸对垂体-甲状腺轴T_SH、T_3、T_4的动态调整程度不一。结论在肾阳虚证动态发展过程中,下丘脑-垂体-甲状腺轴激素水平变化明显且T_SH、T_3、T_4是敏感指标。 Objective To study the dynamic influence of Yougui pill( YP) on hypothalamic-pituitary-thyroid axis in kidney-yang deficiency rats. Methods 90 SD rats were randomly divided into three groups,including the normal group,the model group and YP group. The model of rat kidney-yang deficiency was established by intramuscular injection of hydrocortisone. Rats in the YP group were administrated intragastrically with YP after 8 days,and rats in the normal and model groups were given normal saline. Animals in all groups were sacrificed on 15,23 and 30 days,the levels of TRH,TSH,T_3 and T_4 were measured and the pituitary and thyroid glands were removed, weighed and sectioned for HE staining. Results Compared with the normal group,the levels of TRH,TSH,T_3 and T_4 were decreased significantly in the model group( P〈0.01).The hormone levels of hypothalamus-ituitary-hyroid axis were increased in model rats when treated with YP,in which the pituitary-thyroid hormone levels were enhanced significantly( P〈0.01) with the dynamic difference. Conclusion During dynamic development of kidney-yang deficiency syndrome, the hormone levels of hypothalamus-pituitary-thyroid axis obviously changes and TSH,T_3 and T_4 are sensitive indicators.
