离心法与过滤吸附法处理脂肪组织对脂肪细胞活性影响的比较 被引量:6

摘要 目的比较低速离心法(800 r/min)与过滤吸附法处理脂肪组织对移植脂肪细胞活性的影响。方法从16例健康女性腹部或大腿部获取脂肪后,对脂肪组织分别进行离心(离心组)和过滤吸附(过滤吸附组)处理,应用葡萄糖转运法和锥虫蓝染色法检测细胞活性,并行常规HE染色观察脂肪细胞组织学变化。结果过滤吸附组和离心组葡萄糖转运量分别为(2.00±0.36)mmol/L和(1.50±0.25)mmol/L,差异具有统计学意义(P<0.05)。过滤吸附组和离心组脂肪细胞存活率分别为(91.0±3.0)%和(85.5±3.2)%,差异无统计学意义(P=0.417)。组织学观察显示离心组有少量脂肪细胞存在损伤,细胞膜破裂不完整;过滤吸附组脂肪细胞分布均匀,形态保持完整,基本没有损伤。结论过滤吸附法处理脂肪颗粒与离心法相比,脂肪细胞活性较高,是一种有效且可行的脂肪处理方法。 Objective To compare the effects of the low-speed centrifugation (800 r/min) method and the filtration adsorption method on the viability of autologous fat grafts. Methods Fat tissue was harvested from 16 healthy patients by syringe aspiration. Harvested fat tissue was prepared using two different methods: centrifugation and filtration adsorption, Glucose transport test and trypan blue stain were performed to evaluate the viability of fat grafts. Results The results of the glucose transportation test of the centrifugation group and the filtration adsorption group was (2.00 ±0.36) mmol/L and (1.5 ±0.25) mmol/L, respectively. The viability of lipocyte in filtration adsorption was significantly higher than that in the centrifugation group (P〈0.05). The survival rate of fat is (91.0 ±3.0)% and (85.5 ±3.2)%, respectively. There was no significant difference between the two different methods of fat processing (P=0.417). Conclusion Lipocyte purification by filtration adsorption has a higher viability than centrifugation. It is an efficient and feasible fat processing method.
