广西地方稻种资源核心种质红色种皮全基因组关联分析及鉴定两个新的Rc等位基因 被引量:4

Validation of the Red Pericarp Gene from 419 Rice Landrace Core Collection in Guangxi using Genome-Wide Association Study and Discovery of Two Novel RcAlleles

摘要 稻米种皮颜色可以分为白、红、黑等。在红色稻米种皮中富含原花青素,其具有抗氧化活性,有利于人体健康。本研究以419份广西地方稻种资源核心种质为实验材料,利用全基因组关联分析验证了控制水稻红色种皮的基因Rc。利用In Del标记HM1对97份红色种皮材料的DNA进行PCR,发现C323和C139属于第7外显子有14 bp缺失的红色种皮品种。然后,我们利用Sanger测序技术对这两个自然突变品种的Rc基因进行外显子测序,分析结果表明Rc ^(C139)和Rc ^(C323)属于2种不同的Rc等位基因类型。 Rice can produce white pericarp as well as red, red pericarp and so on. In red rice, the pericarp of the grain contains high content of proanthocyanin, which has antioxidant activity and is beneficial to human health. In the present study, we analyzed the red pericarp of 419 rice landraces in Guangxi by genome-wide association study(GWAS), and validated that the Rc gene regulated the red pericarp trait in rice. We identified two new alleles that arose by natural mutation within the 14 bp deletion cultivars, C139 and C323. The mutation restored the reading frame of the gene, and reverted the bran layer pigmentation to red(wild-type). By sequencing the Rc exons in plants derived from red rice, we were able to demonstrate that mutation within rc resulted in the new, wild-type allele RcC139 and RcC323.
