一种适于SNP芯片分型的玉米种皮组织DNA提取方法 被引量:6

A DNA Extraction Method from Maize Seed Capsule Tissue Suitable for SNP Chip Genotyping

摘要 以京科968、郑单958两套三联体样品为材料,分别采用CTAB和改良CTAB法提取叶片和杂交种种皮的DNA,利用maize SNP384芯片(包括384个SNP位点)进行DNA指纹分析。结果显示:玉米叶片和种皮提取DNA获得基因型数据在成功率、数据质量上一致,均可获得较好的基因型数据;通过将杂交种种皮DNA与母本叶片DNA基因型数据的比较分析,三联体样品亲子关系分析,显示种皮DNA基因型能够代表杂交种母本DNA指纹。因此玉米种皮提取DNA结合SNP芯片检测技术能够在品种权保护和品种鉴定中发挥一定作用。 Using Jingke 968 and Zhengdan 958 two sets triplets as research materials, the DNA was extracted using CTAB and modified CTAB methods from leaves and hybrid seed capsules, and DNA fingerprint was constructed using maize SNP384 chip(including 384 SNP loci). The results showed that good genotype data could be obtained from both leaves and seed capsules DNA, the data success rates and quality were consistent.Comparative analysis of the genotype data from hybrid seed capsules and maternal leaves DNA, and parent-child relationship analysis showed that seed capsules DNA genotype could represent the maternal DNA fingerprint of hybrid. Therefore, the detection technology of maize seed capsules DNA extraction combined with SNP chip could play a certain role in the protection of intellectual property and variety identification.
