慢性阻塞性肺疾病患者外周血CD_4^+CD_(25)^+调节性T细胞和Foxp3的表达 被引量:6

Expression of CD_4^+CD_(25)^+regulatory T cell( Treg) and Foxp3 in peripheral blood of patients with chronic obstructive pulmonary disease

摘要 目的探讨慢性阻塞性肺疾病患者外周血CD_4^+CD_(25)^+调节性T细胞及Foxp3的表达及意义。方法采用病例-对照研究,选取2013年6月至2015年3月在我院就诊的105例慢阻肺患者作为病例组,另选健康人群108例作对照组。对受试者进行肺功能测定;流式细胞术检测外周血Treg表达;ELISA检测血清浓度;RT-PCR检测Foxp3 mRNA表达,并进行相关统计分析。结果与对照组比较,慢阻肺患者FEV_1、FEV_1/FVC水平,IL-10、TGF-β浓度,Foxp3 mRNA及蛋白的表达,CD_4^+Treg、CD_4^+CD_(25)^+Treg的表达和CD_4^+CD_(25)^+Treg所占CD_4^+Treg比例显著降低(P<0.01);Foxp3 mRNA和蛋白与肺功能指标FEV_1(%)和FEV_1/FVC(%)以及蛋白与血清细胞因子IL-4、IL-10、IFN-α、IFN-γ和TGF-β均呈一定的正相关(P<0.05)而与FVC(%)和PEF(%)无显著相关性(P>0.05)。结论慢阻肺患者CD_4^+CD_(25)^+调节性细胞数量减少且血清浓度降低,Foxp3基因表达降低。 Objective To explore the expression of CD4^+ CD25^+ regulatory T cell (Treg) and Foxp3 in periph- eral blood of patients with chronic obstructive pulmonary disease (COPD) and its clinical significance. Methods A total of 105 COPD patients treated in our hospital from June 2013 to March 2015 were selected as the case group and 108 healthy individuals who took examination at the same period were enrolled as the control group. All the subjects underwent pulmonary function testing. The expression of Treg in peripheral blood was detected by flow cytometry. Se- rum concentration was detected by enzyme-linked immuno sorbent assay (ELISA). The mRNA expression of Foxp3 was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results There was significant decrease of PEV1 and PEV1/FVC levels in patients with COPD when compared with the control group (P 〈0.01 ). The expressions of CD4^+ Treg and CD4^+ CD25^+ Treg as well as the percentage of CD4^+ CD25^+ Treg in CD4^+ Treg also decreased significantly while compared with the control group ( all P 〈 0. 01 ). The concentration of IL-10 and TGF-β was also decreased in COPD patients ( P 〈 0.05 ). The expressions of both Foxp3 mRNA and Foxp3 protein in COPD patients were significantly decreased when compared with the control group ( both P 〈 0. 01 ). Both Foxp3 mRNA and Foxp3 protein were positively correlated with FEV1% and FEV1/FVC (%) (P 〈 0.05 ), and Foxp3 mRNA and Foxp3 pro- tein were also positively correlated with serum cytokines IL-d, IL-10, IFN-α, IFN-γ and TGF-β ( all P 〈 0. 05 ). Conclusion The numbers of CD4 + CD25 + Treg cells and serum concentration as well as Foxp3 genetic expression decrease in patients with COPD.
