摘要
目的探讨IL-31在哮喘患者血清、支气管肺泡灌洗液以及支气管组织的表达水平及其与哮喘严重程度的相关性。方法应用荧光酶联免疫法和酶联免疫吸附法(ELISA)测定支气管哮喘患者和健康者血清、支气管肺泡灌洗液以及支气管组织IL-5、IL-13、IL-31以及总IgE的表达水平。结果 (1)支气管哮喘患者血清及支气管肺泡灌洗液IL-31的表达水平明显高于健康者,差异具有显著性(P<0.05);(2)重度哮喘患者支气管组织IL-31的表达水平明显高于中度和轻度患者,差异具有显著性(P<0.05);(3)哮喘血清IL-31的表达水平与IL-5、IL-13、总IgE具有显著正相关,相关系数分别为0.457、0.659、0.783;与第一秒用力呼吸容量(FEV_1)具有显著负相关(r=-0.983,P=0.012);与外周血嗜酸性粒细胞百分比无显著相关性(r=0.756,P=0.087)。结论哮喘患者不同组织的IL-31表达水平能够反映疾病的严重程度,可作为临床监测哮喘的生物指标。
Objective To investigate the expression of IL-31 in asthma patients and its correlation with dis- ease severity. Methods It used fluorescence ELISA and enzyme-linked immunosorbent assay (ELISA) to detect the levels of IL-5, IL-13and IL-31 in bronchoalveolar lavage fluid and bronchial tissues and the expression of total IgE in asthma patients and healthy controls. Results The expression of IL-31 in serum and bronchoalveolar lavage fluid was significantly higher in asthma patients than in healthy controls ( P 〈 0. 05 ), and it was higher in patients with se- vere asthma than in patients with moderate and mild asthma (P 〈 0.05). The serum IL-31 expression was positively correlated with IL-5, IL-13 and total IgE ( t = 0. 457, 0. 659, 0. 783), and negatively correlated with forced expirato- ry volume in one second (FEV1 ) (r = -0. 983, P = 0. 012). It had no correlation with peripheral blood eosinophilic granulocyte (r = 0. 756, P =0. 087). Conclusion The expression of IL-31 in asthmatic patients with different lev- els of the organization can reflect the severity of the disease, which can be used as the biological indicator of clinical monitoring on asthma.
