温阳活血方对慢性心衰大鼠心肌组织转化生长因子β_1的影响

Effect of Warming Yang and Promoting Blood Circulation Decoction on Transforming Growth Factor β1 of Chronic Heart Failure Rats

目的:观察温阳活血方对慢性心力衰竭大鼠心肌组织转化生长因子β1(TGF-β1)的影响。方法:利用肾上腹主动脉缩窄法制备慢性心力衰竭大鼠模型30只随机分为3组各10只,温阴活血方组给予温阳活血方(8m L/kg)每日灌胃,假手术组和模型组给予等量生理盐水灌胃,假手术组只分离服主动脉通不结扎。观察各组大鼠左心室重量指数(LVMI)和胶原容积积分(CVF),RT—PCR测定各组大鼠左心室心肌TGF-β1 mRNA表达水平。结果:与假手术组比较,模型组LVMI、CVF明显升高,左心室心肌TGF-β1 mRNA表达明显升高(P<0.05);与模型组比较,温阳活血方组LVMI、CVF明显下降,左心室心肌TGF-β1 mRNA表达明显下降(P<0.05)。结论:温阳活血方通过影响信号通路中TGF-β1 mRNA的表达而改善心衰大鼠心肌纤维化程度。

Objective ： To observe the effect of WenYang HuoXue Decoction on the expression of TGF-β1 in rats with chronic heart failure.Method： A CHF model of rat was established by constricting the abdominal aorta superior to renal artery.30 male Wistar rats were randomly divided into sham operation group, model group, and WenYang HuoXue Decoction group, 10 rats in each group. WenYang HuoXue Decoction group were given WenYang HuoXue Decoction （ 8kg/kg ） by intragastric administration everyday. Their LVMI and CVF was measured and TGF-β1 mRNA in the left ventricular myocardium were assessed by RT-PCR.Result ： The LYMI, CVF and TGF- β1 mRNA were significantly higher in model group than that in sham operation （ P〈0.01 ） .The LVMI, CVF and TGF-β1 mRNA were significantly lower in WenYang HuoXue group than that of the model group （P〈0.01） .Conclusion： WenYang HuoXue Decoction could significantly improve myocardial fibrosis through decreasing the over-expression of TGF-β1 mRNA.
