摘要
目的 探讨SP对大鼠AP培养细胞中IP3 的水平是否有影响 ,进一步阐述SP产生生物学效应的机制。方法 原代培养成年雌性S -D大鼠AP细胞 48h ,[3 H]-肌醇标记 36h ,加入不同浓度的SP孵育 30min ,超声震碎细胞 ,用AG1-X8阴离子交换层析树脂提取分离 [3 H]-IP ,应用液闪测定样本计数率 (cpm)。 结果 孵育时间为 30min时 ,SP以浓度依赖的方式使AP细胞内IP3 的水平增加。结论 兴奋AP细胞SPR后的生物学效应至少有一部分是通过第二信使IP3 来完成的 ,在信息转导途径上为SP对生殖轴调控的理论进行进一步证明和补充。
Objective This thesis was to determine whether SP had an effect on the levels of IP 3 in primary cultured anterior pituitary cells of the rat.We farther indicated that SP induced the action mechanism of physiology effect. Methods We chose adult female S-D rat AP to culture 48h and label 36h with -inositol and added different concentration of SP for 30min and sonicated the cells .-IP was separated by anion-exchange chromatography using columns of AG1-X 8 resin.The amount of radioactivity was determined with scintillation counter. Results When time course is 30min SP increased the levels of IP 3 in the AP cells in a concentration dependent manner. Conclusion We conclude that biological responses following activation of SPR are mediated in AP cells,at least in part,by the second messenger IP 3 levels.The theory of SP controlling the reproducing axis is confirmed and supplemented in the way of information transforming.
