摘要
目的探讨白细胞介素7受体(IL-7R)基因在膝关节骨关节炎(KOA)疾病中的表达作用及意义,为其诊断和治疗以及进一步研究提供一定的科学依据。方法选取河南大学淮河医院骨科70例患者新鲜滑膜组织,采用Trizol法提取总RNA,采取Applied Biosystems逆转录试剂盒逆转录合成c DNA第一链,用实时逆转录聚合酶链反应(real time RT-PCR)检测膝关节髌上囊滑膜组织中IL-7R基因的表达。结果 IL-7R基因表达在KOA滑膜组织中降低(P<0.05)。结论 IL-7R在KOA滑膜中的表达下降,且与膝关节骨关节炎病变有密切的关联,为检测和治疗KOA疾病提供一定数据支撑和参考。
Objective To discuss expression of knee joint synovial IL-7R genes in knee osteoarthritis (KOA) and the significance so as to provide scientific basis for diagnosis, treatment and research of knee osteoarthritis. Methods Fresh synovial tissue was collected from 70 orthopedic patients in a hospital. Trizol method was used to extract total RNA in the synovial tissue. The Applied Biosystems retrovirus kit was used to synthesize the first chain of eDNA through reverse transcription. The expression of IL-TR gene in osteoarthritic knee synovium was detected by RT-PCR. Results IL-TB gene expression in osteoarthritic knee synovial tissue obviously decreased (P 〈 0.01). Conclusions IL-TR expression in the osteoarthritic knee synovial tissue obviously decreases and is positively correlated with the progression of KOA. Therefore, it provides available information for the detection and treatment of KOA.
