重组腺病毒IL-2诱导HeLa细胞凋亡水平的测定

DETERMINATION OF APOPTOSIS OF HELA CELL INDUCED BY RECOMBINANT ADENOVIRUS-IL2

目的 建立IL - 2基因表达和诱导HeLa细胞凋亡水平的测定方法。方法 用重组腺病毒IL - 2感染HeLa细胞 ,无需DNA提取 ,毛细管电泳直接检测凋亡细胞 ;表达的IL - 2用ELISA和MTT检测。结果 用毛细管电泳能检测出HeLa细胞凋亡峰 ,与环磷酰胺对照一致 ;IL - 2表达量为 2 5ng,表达活性为 70 0IU·ml-1。结论 方法选择性高、重复性好 。

OBJECTIVE To determine the expression of IL-2 and apoptosis of HeLa cell. METHODS HeLa cells were infected by adenovirus-IL2.A capillary electrophoresis method was developed to detect apoptotic cell directly and don't need to extract DNA fragments from cell again. Expression of IL-2 was determined by ELISA and MTT methods.RESULTS Apoptotic peak of HeLacell could be determined by capillary electrophoresis and the results accorded with the model induced by cyclophosphamide. Expressed IL-2 and its bioactivity were 25 ng·ml -1 and 700 IU·ml -1 respectively.CONCLUSION The method is simple,rapid,reliable and good reproducible.
