摘要
目的观察并探讨5种不同灭菌方法、剂量及p H值糖(醇)培养基对不同培养液、不同浓度鼠疫菌的发酵结果,筛选最适鼠疫菌发酵的糖(醇)培养基。方法选取对鼠疫菌具有典型鉴定特征的5种糖(醇),观察制作过程中不同剂量、不同pH值和不同灭菌方法对鼠疫菌发酵结果的影响。结果不同灭菌方法、不同培养液的5种糖(醇)培养基对不同浓度的鼠疫菌发酵结果无影响(141菌株:阿胶糖(+)、鼠李糖(-)、密二糖(-)、麦芽糖(+)、甘油(+);EV 76:甘油(-),其它发酵结果同141菌株,均为正常发酵);不同剂量及pH值的糖(醇)培养基对鼠疫菌发酵结果有一定的影响,特别是剂量为0.5%、pH值为7.2~7.4的阿胶糖和甘油可引起迟缓发酵。结论制备鼠疫菌糖(醇)培养基的最适剂量为1%,pH值为7.6,是鼠疫菌发酵的理想选择值。
Objective To observe and discuss the five different sterilization methods,dosage and pH value of sugar( alcohol)culture medium with different culture,different concentration fermentation result of Y pestis,selected optimum Y pestis fermentation of sugar( alcohol) medium. Methods Selected of Y pestis as identification characteristics of five kinds of typical sugar( alcohol),to observe its influence the result of the Y pestis fermentation in the process of production in different dosage,pH value and sterilization methods conditions. Results The different sterilization methods,different cultures of sugar( alcohol) culture medium with different concentration of Y pestis can not influence the fermentation results(141 strain: arabinose( +),rhamnose(-),dense disaccharide(-),maltose( +),glycerin( +); EV 76 strain: glycerin(-) others fermentation results the same as 141 strain,are all normal fermentation); different dosage and pH value of sugar( alcohol) medium is of certain influence to the result of the plague bacteria fermentation,especially the dosage of 0. 5%,pH 7. 2 ~ 7. 4 gelatin candy and glycerol can cause slow fermentation. Conclusion Reparation of Y pestis sugar( alcohol) medium optimal dosage and pH value were 1% and 7. 6 respectively,is the ideal choice of the Y pestis fermentation.
