蒙药耧斗菜多糖提取方法及质量分数测定的研究 被引量:3

摘要 目的:优选蒙药耧斗菜多糖的提取方法,并测定其质量分数.方法:用回流法与超声法提取蒙药耧斗菜多糖,用硫酸-苯酚法测定其多糖质量分数,设计正交试验优化超声提取法.结果:2种提取方法得到的蒙药耧斗菜粗多糖均为棕色粉末,回流法粗多糖产率为3.47%,多糖质量分数为6.86mg/g,超声法得到的粗多糖产率为2.61%,多糖质量分数为18.90mg/g;正交试验优选出的超声提取条件为:料液比1∶20,超声时间60min,超声温度50℃,超声功率80 W.结论:回流法提取得到的粗多糖产量高,多糖质量分数低,提取时间较长,操作步骤多;超声法得到的粗多糖产量低,多糖质量分数高,提取时间较短,操作步骤较为简便.后者更适合于植物多糖的质量分数测定. Objective:The paper is to optimize the extraction conditions of polysaccharide in Aquilegia viridiflora Pall.,and to determinate its mass fraction.Methods:The polysaccharide was extracted from Aquilegia viridiflora Pall.by reflux extraction and ultrasonic extraction.The mass Fraction was determinated by phenol-sulfuric asid method,and ultrasonic extraction was optimized by the orthogonal test.Results:The brown powder of crude polysaccharide can be obtained fromAquilegiaviridifloraPall.in both methods.By using reflux method,3.47% crude polysaccharide with 6.86mg/g mass fraction of polysaccharides can be detected,while 2.61%crude polysaccharide with 18.90mg/g mass fraction of polysaccharides can be detected in ultrasonic extraction.In addition,the best conditions of ultrasonic extraction are as follows:solid-liquid ratio 1∶20,extraction time 60 min,extraction temperature 50 ℃,and ultrasonic power 80 W.Conclusion:The ultrasonic extraction is much better to detect mass fraction determination of the plant polysaccharide.Contrary to the reflux method,the ultrasonic extraction can obtain less crude polysaccharide and more mass fraction of polysaccharides with less time.
