Effects of Daphne altaica Pall extract on the proliferation,cell cycle and apoptosis of human esophageal carcinoma Eca-109 cells

摘要 目的探讨阿尔泰瑞香正己烷提取物对Eca-109食管癌细胞增殖、细胞周期及凋亡的影响。方法采用噻唑蓝(MTT)法检测阿尔泰瑞香正己烷提取物对Eca-109细胞增殖的抑制作用;流式细胞术检测阿尔泰瑞香正己烷提取物对Eca-109细胞周期的影响;Annexin V-FITC/PE双染法检测阿尔泰瑞香提取物对细胞凋亡的影响。结果阿尔泰瑞香正己烷提取物对Eca-109细胞增殖有抑制作用且具有剂量-时间效应;阿尔泰瑞香提取物阻滞Eca-109细胞于S期,诱导细胞凋亡,药物组细胞凋亡率高于对照组(P<0.05),并且药物组的S期细胞明显高于对照组S期细胞,药物组G1/G2期细胞低于对照组G1/G2期细胞(P<0.01)。结论阿尔泰瑞香正己烷提取物可抑制Eca-109细胞增殖、阻滞细胞周期及诱导细胞凋亡。 Abstract. Objective To evaluate whether n-hexane extract obtained from Daphne altaica Pall has any spe- cific antiproliferative effect to human esophageal carcinoma Eca-109 cells and to study the mechanism of the inhibitory effects. Methods We evaluated cell survival by MTT assay. The esophageal carcinoma Eca- 109 cells apoptosis apoptosis was measured by flow cytometry after Annexin V/PI double staining and cell cycle was measured by flow cytometry after PI single-staining. Results Daphne altaica Pall n-hexane ex- tract exhibited a significant antiproliferative effect on human esophageal carcinoma Eca-109 cells in both time and concentration dependent manners, induces apoptosis and S phase arrest in esophageal carcinoma Eca-109 cells. The apoptosis index of esophageal carcinoma Eca-109 cells treated with Daphne altaica Pall n-hexane extract increased, which were significantly different compared with the control group (P 0.05), esophageal carcinoma Eca-109 cells treated with Daphne altaica Pall n-hexane extract were arrested in S phase in the cell cycle, the number of cells in S phase was increased significantly (P 〈0.01), G1/G2 phase cells reduced dramatically (P 〈0.01). Conclusion This study confirmed that the n-hexane extract had antitumor effect in vitro. The n-hexane extract decreased esophageal cancer cell viability, induced ap- optosis and arrested S phase in esophageal carcinoma Eea-109 cells.
