包头市100例汉族儿童脂联素基因第2外显子单核苷酸多态性与单纯性肥胖的关系 被引量:1

Relationship between the polymorphisms of SNP+45(T→G) in ap M1Exon2 and obesity in 100 Han children of Baotou city

摘要 目的研究脂联素基因(adiponectin gene,ap M1 gene)第2外显子(Exon2)单核苷酸多态性(single nucleotide polymorphism,SNP)与儿童单纯性肥胖的关系,为内蒙古汉族儿童单纯性肥胖的研究与防治提供参考,为疾病的防治提供新的治疗靶点。方法采用整群抽样方法,于2015年9-10月选取内蒙古包头市汉族100例肥胖儿童和100例正常体重儿童。利用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)方法对ap M1Exon2 SNP+45(T→G)多态位点进行基因测定,计算等位基因频率,并且分析其与肥胖代谢的关联性。同时进行身高、体重、血清空腹血糖(FPG)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)的测定。用SPSS 17.0软件进行t检验、χ~2检验。结果肥胖组和正常体重组间FPG、TC、TG、HDL-C和LDL-C水平差异均有统计学意义(P<0.01)。肥胖组和正常体重组ap M1Exon2 SNP+45(T→G)多态位点GG型、TG型、TT型基因型频率分别为14.0%、56.0%、30.0%和4.0%、42.0%、54.0%,基因型频率符合Hardy-Weinberg遗传平衡定律。G、T两组等位基因频率分别为42.0%、58.0%和25.0%、75.0%,两组基因型频率与等位基因频率分布差异有统计学意义(P<0.05)。在肥胖组,SNP+45(T→G)多态位点TG+GG基因型与TT型相比,其FPG、TG、TC、LDL-C水平升高,而HDL-C水平降低,差异均有统计学意义(P<0.05);在正常体重组,TG+GG基因型的FPG、TC较TT型低,差异均有统计学意义(P<0.05)。结论 ap M1Exon2 SNP+45(T→G)单核苷酸多态性与包头市100例汉族儿童单纯性肥胖可能有关。 Objective To study the relationship between the polymorphisms of SNP+45 (T→G) in apM1Exon2 and children obesity, and to provide the reference for preventing and treating obesity in Han children. Methods During September to October in 2015, the cluster sampling method was used to select 100 Han obesity children and 100 Han normal weight children from Baotou city as the subjects. The polymorphisms of apM1Exon2 SNP+45 (T→G) was detected by polymerase chain reaction-restricti0n fragment length polymorphisms(PCR-RFLP) assay. The allele frequencies were calculated, and the correlation of allele frequencies with the obesity metabolism was analyzed. The body height, weight, serum fasting plasm glucose (FPG), total cholesterol (TC), triglyceride(TG), high density lipoprotein-cbolesterol (HDL-C) and low density lipoprotein-cholesterol (LDL-C) were measured for each subject. The data were analyzed statistically with SPSS17.0 software. Results There were significant differences of FPG, TC,TG, HDL-C and LDL-C between obesity group and control group (P〈0.01). The GG, TG and Tr genotype frequencies of apMIExon2 SNP+45 (T→G) in obesity group were 14.0%, 56.0% and 30.0%, respectively; and in normal group were 4.0%, 42.0% and 54.0%, respectively; the genotype frequency was consistent with the Hardy-Weinberg genetic equilibrium law. The G and T allelic frequencies in obesity group and normal group were 42.0%, 58.0% and 25.0%, 75.0%, respectively. There were significant differences of genotype and allelic frequencies for SNP+45 (T→G) between two groups (P〈0.05). In obesity group, as compared with children with TT genotype, the FPG, TG, TC and LDL-C levels enhanced significantly, but HDL-C level reduced significantly in children with TG+GG genotype (P〈0.05). In control group, as compared with children with TF genotype, the FPG and TC levels reduced significantly in children with TG+GG genotype (P〈0.05). Conclusion The polymorphisms of apM1Exon2 SNP+45 (T→G) may be associated with obesity of Chinese Han children in Baotou.
