摘要
Gab蛋白是一类重要的接头蛋白,该家族能与生长因子受体结合蛋白2(growth factor receptor binding protein 2,Grb2)相结合。Gab1是哺乳动物中含量最多、分布最广的Gab蛋白,可通过与各类生长因子、细胞活素类等相互作用来放大和整合信号转导,主要通过RAS-ERK和PI3K-AKT这两条经典途径实现促进细胞生长和分化、控制凋亡、调节免疫等多种生物学功能。研究表明Gab1蛋白与肿瘤的发生、发展以及预后密切相关。全文就Gab1蛋白的结构功能、参与信号转导的作用机制以及在肿瘤中的研究进展进行综述。
Gab proteins,growth factor receptor binding protein 2(Grb2)-associated binder,are important scaffolding adapter proteins. Gab1,the most abundant Gab proteins in mammals,is involved in the amplification and integration of signal transduction evoked by a variety of extracellu-lar stimuli,including various growth factors and cytokines. They are known to play key roles in many biological processes,including cell growth and differentiation,apoptosis and immune regulation,through the two classical signal pathways,RAS-ERK and PI3K-AKT. Some studies have indicated that Gab1 protein is closely related to the occurrence,development and prognosis of tumor.The structure,function and mechanism of action of the Gab1 of scaffolding adapter,with a special focus on its research progress in cancer were reviewed in this paper.
