摘要 目的:探讨乌腺金丝桃与黄芪配伍对心肌缺血再灌注损伤的保护作用。方法:采用冠状动脉左前降支结扎法,建立心肌缺血再灌注损伤模型,测定大鼠血清中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;采用TUNEL法检测心肌缺血再灌注心肌细胞的凋亡;免疫组化法测定心肌缺血再灌注心肌细胞Caspase-3和TNF-α蛋白表达。结果:与M组比较,P组和H+AM1:2组能够明显提高SOD活性,降低MDA含量;与M组比较,H+AM1:2组明显降低大鼠心肌细胞凋亡指数,P组明显降低大鼠心肌细胞凋亡指数;sham组与M组比较,M组心肌细胞中Caspase-3和TNF-α的蛋白量明显增加,与M组比较,H+AM1:2组和P组能够明显降低Caspase-3和TNF-α的蛋白量。结论:乌腺金丝桃与黄芪配伍对缺血再灌注损伤有一定的保护作用。 Objective : To investigate the protective effect of compatibility of Hypericum attenuatum and Astragalus mongholicus on myocardial ischemia reperfusion injury (MIRI). Methods :Myocardial ischemia reperfusion injury models were established by ligation of the left anterior descending artery of the coronary artery. Speroxide dismutase (SOD) activity and malondialdehyde (MDA) content were determined in rats' serum. TUNEL method was used to detect the apoptosis of myocardial cells. Caspase - 3 and TNF - α proteins were determined by immunohistochemistry in myocardial cells of myocardial ischemia and reperfusion. Results: Compared with the model group, the pretreatment group and the H + AM1:2 group could significantly increase SOD activity and reduce MDA content, and H + AM1:2 group could significantly reduce the myocardial apoptosis index in rats. The apoptosis index of myocardial cells was significantly reduced in the pretreatment group. Compared with sham operation group and model group, the protein amount of Caspase - 3 and TNF - α in model group was significandy increased. Compared with the model group, H + AM1:2 group and pretreatment group could significantly reduce Caspase -3 and compatibility of Hypericum attenuatum and TNF-α protein amount. Conclusion:It has a protective effect of Astragalus mongholicus on myocardial ischemia reperfusion injury.
