摘要
目的研究灯盏花素对大鼠肺缺血再灌注损伤后氧化应激的影响,探讨其可能的作用机制。方法通过夹闭左肺门45 min的方法建立大鼠肺缺血再灌注损伤模型,术前30 min分别给予不同浓度灯盏花素(12.5、25、50 mg·kg^(-1))和舒血宁(4 mg·kg^(-1)),并设假手术组和模型组。再灌注2 h后测定肺组织湿/干重比(W/D),比色法测定肺组织中抗氧化酶[超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)]活性和血清中总抗氧化能力(T-AOC)水平、髓过氧化物酶(MPO)活性、丙二醛(MDA)含量;Western blot法测定肺组织NF-κB蛋白表达,TUNEL染色法检测肺细胞凋亡。结果经灯盏花素干预能够显著降低肺缺血再灌注损伤大鼠肺组织W/D、减轻肺水肿,显著提高肺组织中抗氧化酶(SOD、GSH-Px、CAT)活性和血清中T-AOC水平、降低血清中MPO活性和MDA含量,下调NF-κB蛋白表达、降低肺细胞凋亡指数(AI)、改善肺细胞凋亡状况,上述作用均具有一定剂量依赖性。结论灯盏花素可能通过抑制氧化应激反应而对肺缺血再灌注损伤具有一定的保护作用。
Objective To investigate the effect and mechanism of breviscapine (Bre) on the oxidative stress in rats with pulmonary ischemia-reperfusion injury. Methods The pulmonary ischemia-reperfusion rat models were made by clipping left lung door for 45 rain and treated with Bre (12.5, 25 and 50 mg . kg-1) and Shuxuening (4 mg . kg- 1) at 30 min before the operation. A sham operation group and a model group were also established. Two hours after the reperfusion, the wet/dry (W/D) ratio of the lung tissue was examined; the activity of SOD, GSH-Px, and CAT in the lung tissue and MPO in the serum were determined; the level of TAOC and the content of MDA in the serum were determined; the expression of NF-κB in the lung tissue was determined by Western blot, and the apoptosis of lung tissue cells were observed by TUNEL staining. Results The lung tissue W/D ratio of the rats in Bre treated groups was decreased, the activity of SOD, GSH-Px, CAT in the lung tissue and the level of T-AOC were increased; the activity of MPO and the content of MDA were decreased; the expression of NF-κB protein was decreased, and the AI was decreased and the lung cell apoptosis was improved. All the above effects were dose-dependent. Conclusion Bre has protective effect on rats with pulmonary ischemia-reperfusion injury via depressing oxidative stress.
