摘要
[目的]观察雷公藤皮质部和木质部水提物对小鼠毒性反应差异。[方法]将36只雌性ICR小鼠随机分为雷公藤木质部组、雷公藤皮质部组和空白组。分别灌胃给予木质部、皮质部水提物和生理盐水,1次/d,共19 d,期间记录小鼠体重变化情况。末次给药后,心脏取血,检测小鼠丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天冬氨酸氨基转移酶(aspartate aminotransferase,AST)、血尿素氮(blood urea nitrogen,BUN)、血清肌酐(serum creatinine,SCr)、尿酸(uric acid,UA)及血常规;HE染色观察小鼠小肠、肝和肾组织形态学的改变。[结果]与空白组相比,雷公藤木质部组和皮质部组小鼠体重显著降低(P<0.05,P<0.01),出现明显的体征变化,血清ALT、AST、SCr、UA均有不同程度的升高(P<0.05,P<0.01),血常规多项指标发生异常;病理观察显示雷公藤木质部组和皮质部组小鼠的肠壁显著变薄(P<0.05,P<0.01),小肠、肝和肾组织形态均受到了一定程度的损伤,且木质部组更为严重。[结论]雷公藤皮质部和木质部具有相似的肠、肝、肾毒性,可以考虑雷公藤皮质部作为雷公藤提取物制剂原料。
[Objective] To observe the toxic effects of the tripterygium wilfordii cortex and xylem water extract on mice. [Methods] 36 female ICR mice were randomly divided into three groups: tripterygium wilfordii xylem extract group, tripterygium wilfordii cortex extract group and control group, intragastric administration once per day for total 19 days. Record the changes of body weight in mice, and take heart blood to detect alanine aminotransterase(ALT), aspartate aminotransferase(AST), urea nitrogen(BUN), serum creatinine(SCr), uric acid(UA) and routine blood with the last administration; HE staining to observe the liver, kidney and intestine tissue morphology changes. [Results] Compared with the control group, the weight of tripterygium wilfordii group and xylem cortex group of mice decreased significantly(P〈0.05, P〈0.01), there were obvious body sign changes with administration. ALT, AST, SCr, UA were markedly increased(P〈0.05, P〈0.01); Routine blood showed anomalies; Pathological histology observation showed intestinal wall significantly got thinner(P〈 0.05, P〈0.01); Parenchymal cells of intestines, liver, kidney tissues were damaged with various degree, especially for xylem group. [Conclusion] Tripterygium wilfordii cortex and xylem have similar intestines, liver and kidney toxicity, which suggests tripterygium wilfordii cortex can be used as raw material for tripterygium wilfordii preparation
