摘要
目的:探讨miR-146a在慢性免疫性血小板减少症(ITP)患者外周血单个核细胞(PBMC)的表达及其临床意义。方法:选取本院血液科收治的ITP患者35例,选取同期健康者38例作为对照组,比较2组研究对象miR-146a在PBMC和血清中表达量的差异;比较2组血小板计数(PLT)、细胞因子、T细胞亚群的差异;并对ITP患者PBMC中miR-146a的表达量与血清中miR-146a的表达量、PLT、细胞因子、T细胞亚群进行相关性分析。结果:ITP组PBMC中miR-146a的表达量和血清中miR-146a的表达量均显著低于对照组[(0.071±0.004)%∶(0.089±0.008)%,(1.549±0.412)%∶(4.485±0.396)%,均P<0.01]。ITP组外周血中的PLT和Treg显著低于对照组,而IFN-γ、TNF-α、IL-2、Th22显著高于对照组(均P<0.05)。ITP患者PBMC中miR-146a的表达量与血清中miR-146a的表达量、PLT、Treg呈正相关,与IFN-γ、TNF-α、IL-2呈负相关(均P<0.05)。结论:ITP患者PBMC中miR-146a的表达量降低,IFN-γ、TNF-α、IL-2、Th22升高。ITP患者PBMC中miR-146a表达与血清中miR-146a的表达量、PLT、Treg呈正相关,与IFN-γ、TNF-α、IL-2呈负相关。
Objective:To investigate the expression and clinical significance of miR-146 ain peripheral blood mononuclear cell(PBMC)in patients with chronic immune thrombocytopenia(ITP).Method:Thirty-five cases of ITP in Department of Hematology in our hospital were selected,38 healthy persons were selected as the control group.We compared miR-146 aexpressions in PBMC and serum of the two groups.And we compared platelet count,cytokine,T cell subsets in the two groups.We analyzed the correlation between miR-146 aexpression in PBMC and serum levels,platelet count,cytokine and T cell subsets.Result:The expression levels of miR-146 ain ITP patients with PBMC and serum were significantly lower than those in the control group [(0.071±0.004)% vs.(0.089±0.008)%,(1.549±0.412)% vs.(4.485±0.396)%,P〈0.01].The blood platelet count and Treg in the peripheral blood of ITP group were significantly lower than those in the control group,and the IFN-γ,TNF-α,Th22 and IL-2were significantly higher than those in the control group(P〈0.05).The expression level of miR-146 ain ITP patients was positively correlated with the expression of serum,platelet count and Treg,and was negative correlated with IFN-γ,TNF-αand IL-2(P〈0.05).Conclusion:The expression of miR-146 ais decreased but IFN-γ,TNF-α,IL-2and Th22 is increased in PBMC of ITP patients.The expression of miR-146 ain ITP patients is positively correlated with the expression of serum,platelet count and Treg,and is negative correlated with IFN-γ,TNF-αand IL-2.
