环磷酰胺对不同年龄小鼠免疫细胞损伤及修复作用的影响 被引量:5

摘要 目的比较环磷酰胺对幼年鼠、老年鼠与成年鼠免疫系统的影响。方法雄性C57BL/6幼年、成年、老年鼠各32只,各年龄小鼠均随机分为对照组(12只)和模型组(20只)。模型组成年鼠、老年鼠腹腔注射环磷酰胺100 mg/kg,幼年鼠腹腔注射环磷酰胺72 mg/kg。对照组腹腔注射等剂量生理盐水。分别在给药后第3、7、14、28天采用流式细胞仪、血细胞计数仪检测各年龄3只对照组、5只模型组小鼠相关免疫指标。结果给药后,与成年鼠相比,幼年鼠胸腺CD8+T细胞、CD4+CD8+T细胞、血浆Ig A、Ig G损伤恢复情况更好(P<0.05);脾脏Treg细胞恢复情况较差(P<0.05)。与成年鼠相比,老年鼠脾脏指数下降更明显(P<0.05);外周血NK细胞、B细胞、中性粒细胞、CD4+T细胞恢复情况较差(P<0.05);白细胞数下降更明显且预后较差(P<0.05)。结论应用环磷酰胺后,幼年鼠免疫系统损伤、恢复情况与成年鼠基本一致。环磷酰胺对老年鼠免疫系统的损伤较成年鼠更严重且预后较差。 Objective To compare the effect of immune system injury induced by Cyclophosphamide among juvenile, adult and aged mice. Methods The 32 male C57BL/6 mice of every age were randomly divided into control group and model group. Every age control group had 12 mice and model group had 20 mice. Adult and aged mice were treated with Cyclophosphamide 100 mg/kg by intraperitoneal injection. Juvenile mice were treated with Cyclophosphamide 72 mg/kg by intraperitoneal injection. And the control mice were treated with saline in the same way as the Cyclophosphamide treated mice. Some immune indexes were measured by flow cytometer and blood-counter system at 3, 7, 14, 28 days after treatment. 3 mice of control group and 5 mice of model group of every age were measured every time. Results Compared with the aduh mice, the proportion of thymus CD8^+T cells, CD4^+TCD8^+T cells, plasma IgA, IgG had better prognosis (P 〈 0.05), but spleen Treg cells had poor prognosis (P 〈 0.05). Compared with the adult mice, spleen indexes of aged mice were reduced more seriously (P 〈 0.05). The percentage of peripheral blood NK cells, B cells, neutrophils and CD4^+T cells had poor prognosis (P 〈 0.05); the number of white blood cell were reduced more seciously and it had poor prognosis (P 〈 0.05). Conclusion After treatment with Cyclophosphamide, the damage and repair of juvenile mice's immune system are consistent with the adult mice's, but the damage and repair of aged mice's are more severe.
