摘要
为了探讨近红外光谱分析技术作为快速分析技术应用于检测万古霉素血药浓度的可行性,配制浓度梯度在2~98μg/m L的万古霉素绵羊血浆样品,采用透射模式采集样品在10 000~4 000 cm-1的透射光谱。利用K-S方法将样品集划分为校正集和验证集,通过预处理方法的优化选择最优的光谱预处理方法,建立万古霉素血药浓度分析模型并对模型进行评价。利用一阶导数Norris平滑对光谱进行预处理后建立的PLS模型,模型的RMSEP=3.182μg/m L,Rp=0.993 9,RPD=8.6,可用于血浆中万古霉素的定量分析。近红外光谱分析技术用于血浆中万古霉素浓度具有很高的准确性,可用于体外血浆样品中万古霉素浓度的检测。
To investigate the feasibility of using near infrared spectroscopy as a quick analysis tool to detect the concentration of vancomycin in plasma, different concentrations of vancomycin sample ranging from 2 to 98 μg/mL in plasma of sheep were prepared. Then,near infrared transmission spectra ranging from 10 000 cm-1 to 4 000 cm-1 of the samples were collected. The data were divided into calibration set and validation set using K-S method. After choosing appropriate pretreatment methods, models of vanco- mycin concentration in sheep plasma were established and evaluated. A PLS model was established after being pretreated by Norris smoothing, of which the value of RMSEP was 3.182 μg/mL, Rp was 0.993 9 and RPD was 8.6. That means that the model has satis- fying forecast precision and can be used to qualify vancomycin in blood. Near infrared spectroscopy can predict the concentration of vancomycin in blood precisely, so it can be used to quickly detect the concentration of vancomycin in blood in vitro.
