摘要
目的观察双歧杆菌对大鼠肠上皮細胞Toll样受体2(TLR2)和Toll样受体4(ILR4)表达的影响,为双歧杆菌治疗新生儿坏死性小肠结肠炎(NEC)提供理论依裾。方法选取50只健康SD大鼠,适应性喂养1周后采取TNBS法造模,将以上大鼠随机分为对照组与观察组,每组25只。对照组给予常规饲养,观察组在常规饲养的基础上给予双歧杆菌经口2 ml/d灌服,连续饲养3周后比较两组大鼠一般状况、肠黏膜大体评分、组织学评分与RTPCR检测结果。结果观察组大鼠一般状况总体要比对照组大鼠轻,且IOd后有缓解迹象;观察组大鼠肠黏膜大体评分、组织学评分低于对照组,大鼠肠组织中TLR2与TLR4表达高于对照组,差异有统计学意义(P<0.01)。结论双歧杆菌能够抑制TLR2与TLR4的表达,减轻肠黏膜损伤程度,促进肠道正常生理功能的恢复,为双歧杆菌治疗NEC提供了实验支持。
Objective To observe the effect of bifidobacterium on the expression of toll - like receptor 2 (TLR2) and toll - like receptor 4 ( TLR4) in intestinal epithelium of rats, and to provide theoretical basis for the treatment of necrotizing enterocolitis ( NEC) by bifidobacterium. Methods Fifty healthy SD rats were selected. TNBS was used to make the model after 1 week of adaptive feeding. The rats were randomly divided into control group and observation group, 25 rats in each group. The control group was given conventional feeding, the observation group was fed with bifidobacteria 2 ml/d on the basis of routine feeding, and the general condition, gross score of intestinal mucosa, histological score and RT - PCR results were compared between the two groups after 3 weeks of continuous feeding. Results The general condition of the observation group was lighter than that of the control group, and there was remission after 10 days. The gross score of intestinal mucosa and histological score of the observation group were lower than those of the control group, the expression of TLR2 and TLR4 in the intestinal tissue was significantly higher than that in the control group (P 〈0. 01). Conclusion Bifidobacterium can inhibit the expression of TLR2 and TLR4, reduce the degree of intestinal mucosal injury, and promote the recovery of normal physiological function of the gut. This study provides experimental support for the treatment of NEC by bifidobacteria.
