结直肠癌组织中NGFR基因启动子甲基化的研究 被引量:2

摘要 目的检测结直肠癌组织中NGFR基因启动子区的甲基化状态及其表达水平,探讨其临床意义。方法收集江西省人民医院2015年9月至2016年3月手术切除的91例结直肠癌组织及其相应的癌旁组织标本作为研究对象,通过MSP及RT-PCR法检测NGFR启动子区的甲基化状态及m RNA表达水平,分析结直肠癌组织中NGFR启动子区甲基化水平与临床资料的相关性。结果与癌旁组织相比,结直肠癌组织中NGFR的表达水平降低且伴随启动子区高甲基化。甲基化水平与TNM分期、分化程度及肿瘤浸润深度相关(P<0.05),随着肿瘤分化程度的降低、TNM分期的提高及浸润深度的增加而具有更明显的甲基化水平。结论 NGFR启动子区高甲基化可能在结直肠癌的发生过程中发挥重要作用。 Objective To detect the expression of NGFR and the methylation status of its gene promoter region in colorectal cancer,so as to analyze their correlation. Methods 91 patients with of colorectal cancer tissues and adjacent normal tissues were collected from September 2015 to March 2016 in Jiangxi People's Hospital as research subjects. The NGFR m RNA expression and the methylation status of NGFR gene promoter region in colorectal cancer were detected by using RT-PCR and MS-PCR respectively. In addition,the correlation of NGFR gene methylation status with clinicopathological parameters was analyzed. Results In comparison with adjacent normal tissues,colorectal cancer tissues displayed a low expression level of NGFR m RNA and a high methylation level of gene promoter region. Besides,the methylation level was significantly correlated with tumor invasion depth,differentiation and TNM staging(P〈0.05). With deep invasion,poor differentiation or higher TNM staging,methylation level of gene promoter region was more significant in colorectal cancer tissues. Conclusion High methylation of NGFR gene promoter region may be a significant contributing factor leading to the development of colorectal cancer.
