摘要
目的研究维生素D(Vit D)是否可通过抑制脓毒症小鼠机体过度的炎症反应来减轻机体免疫损伤及增加存活率。方法将雄性清洁健康ICR小鼠30只按随机数字表法均分为正常对照组,脓毒症组(LPS组)和维生素D(Vit D)干预组。腹腔注射脂多糖(LPS 5mg/kg)制备大鼠脓毒症模型,干预组在制备脓毒症模型前给予维生素D3(1000IU/kg)灌胃注入,连续7d,采用酶联免疫吸附试验(ELISA)检查血清促炎因子(IL-6、TNF-α)及血清25(OH)D的水平;观察各组小鼠3d生存率。结果注射LPS后,Vit D干预组血清促炎因子IL-6[(152.51±16.61)ng/L]比LPS组[(378.42±54.21)ng/L]降低,血清TNF-α[Vit D干预组:(398.38±64.51)ng/L]水平较LPS组[(982.59±132.84)ng/L]减低,均有统计学差异(P<0.05);Wit D干预组血清25(OH)D[(37.52±5.56)mg/L]水平较LPS组[(18.85±6.14)mg/L]显著升高(P<0.05),但两组均较正常对照组[(52.38±8.56)mg/L]明显降低,具有显著差异(P<0.05);正常对照组3d生存率为100%,LPS组3d生存率为10%,Vit D干预组3d生存率为50%,与LPS组比较,Vit D干预组3d生存率显著升高(P<0.05)。结论 Vit D可通过抑制抗炎因子的分泌,抑制脓毒症小鼠机体过度的炎症反应,进而显著降低死亡率。
Objective To study on Vitamin D(Vit D) the effects of inhibit the excessive inflammatory response sepsis in mice to ease the body's immune injury and increase the survival rate. Methods 30 clean and healthy male ICR mice according to random number table method were divided into normal control group,sepsis group(LPS) and vitamin D(Vit D) intervention group. Rat model of sepsis made by intraperitoneal injection of lipopolysaccharide(LPS 5mg/kg),Intervention group:rat given vitamin D3 before preparation of sepsis model,(1000iu/kg) by gastric injection for seven days in a row,Using enzyme-linked immunosorbent assay(ELISA) to check serum proinflammatory factor(IL-6,TNF alpha) and serum 25(OH) D levels;Observing 3d survival rate of mice in 3 groups. Results After injection of LPS,serum proinflammatory factor IL-6(152.51±16.61)ng/L in Vit D intervention group was lower than the one LPS group(378.42±54.21)ng/L,The serum TNF alpha [Vit D intervention group:(398.38±64.51ng/L) level was lower than the LPS group(982.59±132.84)ng/L. Both were statistically different(P〈0.05);serum 25(OH) D((37.52±5.56)mg/L] level in Wit D intervention group increased significantly(P〈0.05),compared with serum 25(OH) D in LPS group(18.85±6.14)mg/L. But the two groups significantly decreased compared with normal control group(52.38 ±8.56)mg/L,which had significant difference(P〈0.05); Normal control group 3d survival rate was 100% in 3 days,LPS group(3DS) rate 10%,Vit D intervention group(3DS) rate 50%.Compared with LPS group,Vit D intervention group 3DSR significantly increased(P〈0.05). Conclusion Vit D can inhibit the antiinflammatory factor secretion and body excessive inflammation of sepsis mice and,therefore,reduce the mortality significantly.
