铁树雄花提取物对鼻咽癌的抑制作用研究 被引量:1

摘要 目的观察铁树雄花水提和水提醇沉提取物对荷鼻咽癌裸鼠抑制作用。方法对铁树雄花分别水提和水提醇沉(醇沉)进行提取有效成份,采用鼻咽癌细胞接种裸鼠待长出实体瘤,将实体瘤移植裸鼠皮下建立荷鼻咽癌裸鼠模型,用铁树雄花提取物(水提取物、水提醇沉提取物)分别给实验组裸鼠灌胃0.3ml/10g,对照组给饮用水,阳性对照组给予去氧氟尿苷,每日1次,连续30d。每周测量体质量和肿瘤大小2次,30d给药后处死小鼠,解剖称肿瘤重量,体积计算公式V=a*b*c*π/6,计算抑瘤率,对肿瘤行组织免疫组化(TUNEL)进行凋亡检测,评价铁树雄花提取物(水提取物、醇沉提取物)对裸鼠荷瘤的抑制作用。结果醇沉组对肿瘤的抑制作用明显,与阴性对照组和水提组比较均有统计学差异(P<0.05);水提组对肿瘤抑制与阴性对照组有统计学差异(P<0.05),但抑制作用没有醇沉组明显;TUNEL结果显示,铁树雄花提取物对肿瘤的凋亡具有很大促进作用,与动物实验结果相吻合。结论铁树雄花水提和醇沉提取物对荷鼻咽癌裸鼠肿瘤具有抑制作用,为铁树雄花开发成抗肿瘤药物研究提供了实验基础。 Objective Study inhibitory effect of Sago Cycas male flower water and water extract-alcoho on Nasopharyngeal Carcinoma. Methods Extracts active ingredient of Sago Cycas male Flower by water and water extract-alcoho,Nasopharyngeal Carcinoma cell inoculated into nude mice to solid tumor,Establishment of nude mice model of nasopharyngeal carcinoma by Nasopharyngeal Carcinoma cell inoculated into nude mice to solid tumor and transplanted in the subcutis of nude mice. Water and Ethanol extracts gavage nude mice dose is 0.3ml/10 g in experience group,Control group gavage water,Positive group gavage doxifluridine. Measure the body and tumour weight twice on week,the mice were killed to collect the samples of tumou on 30 days.Weigh and Calculation volume(V=a*b*c*π/6)for the rate of inhibitory. Evaluate inhibitory effect by TUNEL of tumou. Results Ethanol extracts has obvious inhibitory effect compared with the control group and water extraction group,the difference was statistically significant(P0.05). Water extraction group compared with the control group,the difference was statistically significant(P0.05),but the inhibition effect was lower than Ethanol extracts group. The result of TUNEL is the same as animal experiments,Sago Cycas male flower water and ethanol extracts has a significant role on inhibitory of Nasopharyngeal Carcinoma. Conclusion Sago Cycas Male Flower water and ethanol extracts has a significant role on inhibitory of Nasopharyngeal Carcinoma,It provides the experimental basis for Sago Cycas male flower developing anti-anticancer drugs.
