摘要
本试验应用过碘酸雪夫染色法、荧光定量PCR、间接酶联免疫吸附试验对饲喂双歧杆菌后雏鸡肠道杯状细胞数量、MUC2 mRNA表达量及肠液黏蛋白2(MUC2)含量变化进行研究,揭示双歧杆菌对雏鸡局部黏膜免疫的影响。结果发现,饲喂双歧杆菌后1~18 d雏鸡肠道杯状细胞数量、MUC2 mRNA表达量及肠液MUC2含量不同程度高于对照雏鸡。其中,回肠杯状细胞数量、MUC2 mRNA及肠液MUC2含量于饲喂后1~4 d明显高于对照雏鸡(P〈0.05或P〈0.01);结直肠MUC2mRNA于饲喂后1~7 d极显著高于对照雏鸡(P〈0.01)。结果表明,双歧杆菌能增加雏鸡肠道杯状细胞数量,提高MUC2的分泌量,增强雏鸡肠道黏膜免疫功能。
In order to study the effects of Bifidobacterium on local mucosal immune,periodic acid-schiff staining,quantitative real-time PCR and indirect enzyme-linked immunosorbent assay were used to study the number of goblet cells,expression of MUC2 mRNA of chicks’ intestinal tract and the content of MUC2 in intestinal mucus after administrated Bifidobacterium. The results were as follows: the number of goblet cells of chicks’ intestinal tarct,expression of MUC2 mRNA of chicks’ intestinal tract and the content of MUC2 in intestinal mucus in chicks administrated with Bifidobacterium were significantly enhanced when compared with those in the control group. Meantime,the number of goblet cells of ileum intestinal tract,the impression of MUC2 mRNA of ileum intestinal tract and the content of MUC2 in ileum intestinal mucus were also significantly higher than those in the control group at 1- 4 days after administrated Bifidobacterium( P 〈 0. 05 or P 〈 0. 01); the impression of MUC2 mRNA of colon intestinal tarct is extremely significantly higher than the control group in 1- 7 days after administrated Bifidobacterium( P 〈 0. 01). The results showed that Bifidobacterium can enhance the number of goblet cells and affect the seretion of MUC2,each of which can enhance chicks’ intestinal mucosal immune function.
