小鼠急性肝损伤中谷胱甘肽S转移酶A1的表达分析 被引量:2

摘要 为研究谷胱甘肽S转移酶A1(GSTA1)在3种急性肝损伤动物模型中的表达情况,选用雄性昆明系小鼠32只,随机分为4组,包括CCl_4组、APAP组、酒精组和对照组,按本课题组前期建立的方法复制三种急性肝损伤模型。采集血清检测转氨酶(ALT、AST),肝脏检测肝组织指标(SOD、MDA、GSH、GSH-Px),应用RT-PCR方法测定肝组织中GSTA1 mRNA表达量。结果显示,3种模型组中血清转氨酶及肝组织指标与对照组相比差异极显著(P<0.01);3种模型组中GSTA1 mRNA的表达量与对照组相比极显著降低(P<0.01),其中CCl_4组降低了4.9倍、APAP组降低了2.1倍、酒精组降低了3.7倍。研究表明,在急性肝损伤中,GSTA1 mRNA的表达水平与肝损伤的程度呈负相关。 To analyze the expression of glutathione S-transferase A1( GSTA1) in three acute hepatic injury models,32 male Kunming mice were randomly divided into 4 groups,CCl4,APAP,alcohol and control. Three hepatic injury models were replicated according to the methods established by our research group. Serum levels of transaminases( ALT and AST) and liver homogenate indicators( SOD,MDA,GSH and GSH-Px) were tested and GSTA1 mRNA expression was determined by RT-PCR. The results showed that transaminases serum levels and liver homogenate indicators were significantly changed( P 〈 0. 01),and GSTA1 mRNA expression was significantly decreased( P 〈 0. 01),which were by 4. 9 times( CCl4model),2. 1 times( APAP model) and 3. 7times( ethanol model). These results indicated that mRNA expression levels of GSTA1 in liver are negatively correlated with the degree of hepatic injury.
