高脂饮食对孕鼠子痫前期样改变及神经元mGluR1影响研究 被引量:3

摘要 目的探讨高脂饮食对孕鼠子痫前期样改变及神经元m GluR1的影响。方法将40只雌性大鼠随机分为非孕对照组(NN组)、妊娠对照组(PN组)、非孕高脂饮食组(NF组)及妊娠高脂饮食组(PF组),每组各10只。测量4组雌性大鼠收缩压、尿蛋白及血清生化指标;RT-PCR、Western Blot检测代谢型谷氨酸受体(m GluR1)mRNA及蛋白表达。结果孕鼠妊娠第16、20天,PF组收缩压明显高于NN组、PN组、NF组(P<0.05)。孕鼠妊娠第15、19天,PF组24 h尿蛋白含量明显高于NN组、PN组、NF组(P<0.05)。NF组TG、LDL-C、FFA、Lp-PLA2、LDL-C/HDL-C明显高于NN组,HDL-C明显低于NN组(P<0.05)。PF组TG、LDL-C、FFA、Lp-PLA2、LDL-C/HDL-C明显高于PN组,HDL-C明显低于PN组(P<0.05)。PF组TG、LDL-C、FFA、Lp-PLA2、HDL-C明显高于NF组(P<0.05)。RT-PCR及Western blot检测显示,PF组m GluR1 mRNA及蛋白表达明显增强,高于NN组、PN组、NF组(P<0.05)。结论高脂饮食可诱发孕鼠子痫前期样改变,m GluR1 mRNA及蛋白表达上调可促进子痫发生。 Objective To investigate high-fat diet on pregnant rats induced preeclampsia-like changes and neurons m GluR1 impact.Methods A total of 40 female rats were randomly divided intothe non-pregnant nutritious group( NN group),pregnantnutritious group(PN group),non-pregnant with high-fat diet feeding group(NF group)and pregnant with high-fat diet feeding group(PF group),there were 10 rats in each group. The systolic pressure,urine protein and serum biochemical indexes of four groups were measured;using RTPCR,Western Blot to inspect mRNA and protein expression in metabolic glutamic acid receptor( m GluR1). Results On the 16 th and20th pregnant day,the systolic pressure in PF group was significantly higher than that in the other three groups( P〈0. 05). On the15 th and 19 th pregnant day,the 24 hours urine protein level in PF group was significantly higher than that in the other three groups(P〈0. 05). Compared with NN group,TG,LDL-C,FFA,Lp-PLA2 and LDL-C/HDL-C were significantly higher,while HDL-C was significantly lower in NF group( P〈0. 05). Compared with PN group,TG,LDL-C,FFA,Lp-PLA2 and LDL-C / HDL-C were significantly higher,while HDL-C was significantly lower in PF group( P〈0. 05). Compared with NF group,TG,LDL-C,FFA,Lp-PLA2 and HDL-C were significantly higher in PF group( P〈0. 05). The result of RT-PCR and Western blot inspection showed that the expressions of mRNA and protein of m GluR1 in PF group were significantly increased and higher than those in the other three groups( P〈0. 05). Conclusion High-fat diet may result in preeclampsia-like changes of pregnant rats,the up-regulated expressions of mRNA and protein of m GluR1 may promoteeclampsia happened.
