黄芪提取物对病毒性心肌炎细胞模型的治疗作用 被引量:4

The Effect of Astragalus Extract on Cell Model of CVB3-induced Virus Myocarditis

摘要 目的本文旨在研究黄芪提取物(Astragalus extract,AE)对CVB3病毒诱导的病毒性心肌炎细胞模型的保护作用,为进一步研究提供理论依据。方法利用CVB3病毒和原代心肌细胞建立病毒性心肌炎细胞模型并使用AE对模型进行干预,心肌细胞活性、培养基中LDH、CK-MB水平被检测以对AE的干预效果进行评价。通过检测AE对病毒增殖的抑制作用,细胞内凋亡相关基因Fas、心肌功能相关基因C-Myc和TNF-α转录水平的变化对AE的作用机制进行研究。结果与病毒组比较,AE能够显著的抑制由CVB3诱导的心肌细胞活性减弱和降低培养基中心肌酶水平(P<0.01);AE能够显著的抑制CVB3病毒的增殖,与模型对照组比较,AE对CVB3诱导的Fas、C-Myc和TNF-α基因转录水平的增加有显著的抑制作用(P<0.01)。结论AE对CVB3病毒诱导的病毒性心肌炎细胞模型具有一定的保护作用,这种保护作用与抑制病毒增殖及影响Fas、C-Myc和TNF-α基因表达水平有关。 Objective The purpose of this study was to investigate the effect of Astragalus Extract (AE) on cell model of CVB3-induced Virus Myocarditis, which will provide a theory basis on advanced research. Methods Cell model of Virus Myocarditis was prepared with CVB3 virus and primary cardiomyocytes, which then was treated with AE. Therapeutic effects of AE on cell model of CVB3-induced Virus Myocarditis were evaluated by detecting cell viability with MTT kit, LDH and CK-MB level in Cell culture medium with ELISA. Inhabiting effect of AE on CVB3 virus proliferation and decrease gene express, including Fas, C-Myc and TNF-u were detected to study protective mechanism of AE on CVB3-induced Virus Myocarditis. Results Comparing with Virus group, AE could significantly increase cell viability and decrease myocardial enzyme level (P〈0.01). Fas, C-Myc and TNF-α mRNA level were significantly increased in model group, which were inhabited by AE (P〈0.01). Conclusion AE had certainly protective effect on CVB3-induced Myocarditis, which was related to inhabit virus proliferation and affect Fas, C-Myc and TNF-α gene express.
