柴胡疏肝散对肝气郁结证大鼠前额叶及海马单胺类神经递质的影响 被引量:6

Effect of Chaihu Shugan Power on levels of monoamine neurotransmitters in the frontal cortex and hippocampus in rats with liver-qi stagnation syndrome

摘要 目的研究柴胡疏肝散对肝郁证大鼠行为学和大鼠前额叶及海马单胺类神经递质的影响。方法 30只大鼠随机均分为空白对照组(对照组)、肝郁证模型组(模型组)、柴胡疏肝散组(治疗组)3组。用慢性束缚应激结合孤养法建立肝郁证大鼠模型,测评开野、蔗糖水实验,采用高效液相色谱法测定大鼠前额叶及海马单胺类神经递质。结果模型组大鼠造模成功,与对照组比较,前额叶皮质及海马中的5-HT、5-HIAA、DA含量均下降,NE含量呈增加趋势,但差异无统计学意义;治疗组与模型组相比,5-HT、5-HIAA与DA含量均增加,差异有统计学意义(P<0.05,P<0.01),NE含量呈减少趋势,但差异无统计学意义(P>0.05)。结论柴胡疏肝散具有治疗肝郁证的作用,其作用机制可能与其能够调节肝郁证大鼠脑5-HT、5-HIAA、NE、DA有关,主要是通过增加海马中的5-HT、5-HIAA、DA水平,从而有效发挥抗肝郁作用。 Objective To study the effect of Chaihu Shugan Powder on the ethological changes and levels of monoamine neurotransmitters in the frontal cortex and hippocampus in rats of liver qi stagnation syndrome. Methods 30 rats were equally divided into the normal group( control group), liver stagnation syndrome model group( model group) and Chaihu Shugan Powder group( treatment group). The rat model of liver stagnation syndrome was established by chronic restraint stress and keeping them alone. Each rat was reviewed by Open-field test,Sucrose test and the levels of monoamine neurotransmitters in the frontal cortex and hippocampus were determined by HPLC. Results The model group was created successfully,and compared with the control group,the levels of 5-HT,5-HIAA,and DA in the frontal cortex and hippocampus were decreased,while the level of NE tend to be increased,but there was no statistical significance in difference. But to compare the treatment group and the model group,the levels of 5-HT,5-HIAA,and DA in the frontal cortex and hippocampus were increased,and the differences were statistically significant( P 〈 0. 05,P 〈 0. 01),the level of NE tend to be decreased,but there was no statistical significance in difference( P〉 0. 05). Conclusion Chaihu Shugan Powder has a therapeutic effect on liver stagnation syndrome and the action mechanism may be related with its adjustment of the levels of 5-HT,5-HIAA,NE,DA in the brain of rats with liver stagnation syndrome,mainly by increasing the levels of 5-HT,5-HIAA,DA in hippocampus so as to effectively resist liver stagnation.
