具有荚膜的白假丝酵母菌蛋白质组学分析

Proteomic Analysis of Candida Albicans with Capsule

目的:研究从淋病患者阴道分泌物中分离鉴定的具有荚膜的白假丝酵母菌蛋白质组学特征。方法:采用二维凝胶电泳(2-DE)技术分离从淋病患者阴道分泌物中分离鉴定的具有荚膜的白假丝酵母菌(C1-4)和无荚膜的白假丝酵母菌(ATCC14053)的菌体蛋白质,用Image Master 2D platinum 7.0软件识别差异蛋白点,MALDI-TOFMS鉴定差异蛋白。结果:C1-4与ATCC14053相比,显著差异表达蛋白点232个,其中180种蛋白表达明显上调,52种蛋白表达明显下调,质谱鉴定了32差异蛋白质点,8个得到鉴定,均在C1-4菌株显著表达。结论:具有荚膜的白假丝酵母菌与无荚膜的白假丝酵母菌菌株相比,蛋白质表达存在明显差异。

Objective： To study the proteomic characteristics of candida albicans with capsule isolated from vaginal secretions of gonorrhea patients. Methods： Two-dimensional gel electrophoresis（ 2-DE） was performed to separate proteins from candida albicans with capsule（ C1-4） isolated from vaginal secretions of gonorrhea patients and the standard strain of candida albicans with no capsule（ ATCC14053） respectively. Image Master 2D platinum 7. 0 software was used to analyze 2-DE images and the differential protein spots between C1-4 and ATCC14053 were identified by MALDI-TOF-MS.Results： Campared with ATCC14053,232 differential protein spots in C1-4 were detected; 180 proteins were significantly higher than that in ATCC14053; 8 proteins of which were identified by MALDI-TOF-MS; 52 proteins were significantly lower than that in ATCC14053. Conclusion： The protein expression of candida albicans with capsule was significantly different campared with candida albicans with no capsule.
