摘要
目的探讨细粒棘球蚴(Echinococcus granulosus,E.g)感染对小鼠肝脏细胞周期相关蛋白表达的影响。方法C57BL/6J小鼠16只随机分为实验组和对照组,实验组8只以细粒棘球蚴原头节混悬液直视下注射肝脏,对照组8只以相同方法注射无菌PBS,感染180 d后处死小鼠,检测血清中转氨酶的变化,HE染色后观察肝组织病理改变,采用Western blot检测p-ERK、PCNA、cyclin A、cyclin D1和cyclin E在肝内的表达。结果与对照组相比,实验组小鼠血清中ALT和AST分别升高0.34倍和2.33倍;镜下可见肝脏病灶出现坏死、炎性细胞浸润和纤维组织形成等多种病理改变,肝内PCNA蛋白表达增加,且在两组间的表达差异有统计学意义(t=2.37,P<0.05)。结论 E.g感染宿主后,肝脏损伤与感染应激条件下的代偿性增生同时存在。
Objective To investigate the effect of infection with Echinococcus granulosus on cell cycle-related proteins expression in livers of C57BL/6J mice. Methods C57BL/6J mice were randomly divided into experimental(8) and control(8) group. In the experimental group, mice were infected by intra-hepatic injection of protoscolex suspension of Echinococcus granulosus after laparotomy, while in the control group, mice were injected with sterile PBS in the same way. The mice were euthanized at 180 days, to determine alanine aminotransferase(ALT) and aspartate aminotransferase(AST) changes in serum. After collecting liver samples, pathological alterations were observed by haematoxylin-eosin staining. The protein expressions of p-ERK, PCNA, cyclin A, cyclin D1, and cyclin E in livers were analyzed by Western-blot. Results In comparison with the control group, the levels of ALT and AST in the experimental group increased to 0.34 folds and 2.33 folds, respectively. Pathological changes were found in the liver lesions, including necrosis, inflammatory cells infiltration and the formation of fibrous tissues. Western-blot showed the protein expressions of PCNA was significantly increased(t=2.37, P〈0.05). Conclusions After the process of Echinococcus granulosus infection, hepatic injury exists as long as compensatory liver regeneration under the condition of infection stress.
