虫草素调节肺癌细胞株A549恶性生物学行为的实验研究 被引量:3

摘要 目的:研究虫草素对肺癌细胞株A549增殖、凋亡、侵袭相关分子表达量的影响。方法:培养肺癌细胞株A549,用不同剂量虫草素(0、0.25、0.5、1.0、2.0、4.0ng/mL)处理24h后,测定细胞中增殖、凋亡、侵袭相关分子的mRNA表达量。结果:0.25、0.5、1.0、2.0、4.0ng/mL虫草素处理后,Caspase-3、Caspase-8、NOX1、LATS1的mRNA表达量均显著高于0ng/mL虫草素,CyclinD1、Bcl-2、c-Myc、cFLIP、TRAF6、N-cadherin、Vimentin的mRNA表达量均显著低于0ng/mL虫草素;虫草素剂量越大,Caspase-3、Caspase-8、NOX1、LATS1的mRNA表达量越高,CyclinD1、Bcl-2、c-Myc、c-FLIP、TRAF6、N-cadherin、Vimentin的mRNA表达量越低。结论:虫草素对肺癌细胞株A549中促凋亡基因的表达具有促进作用,对促增殖和促侵袭基因的表达具有抑制作用。 Objective:To study the effect of cordycepin on proliferation,apoptosis and invasion-related molecule expres-sion in lung cancer cell lines A549.Methods:Lung cancer cell lines A549 were cultured and treated with different doses of cordycepin (0,0.25,0.5,1.0,2.0 and 4.0 ng/mL)for 24 h,and then the proliferation,apoptosis and invasion-related mole-cule mRNA expression in cells were detected.Results:After 0.25,0.5,1.0,2.0 and 4.0 ng/mL cordycepin treatment, Caspase-3,Caspase-8,NOX1 and LATS1 mRNA expression were significantly higher than those after 0 ng/mL cordycepin treatment while CyclinD1,Bcl-2,c-Myc,c-FLIP,TRAF6,N-cadherin and Vimentin mRNA expression were significantly lower than those after 0 ng/mL cordycepin traetment;the greater the cordycepin dosage,the higher the Caspase-3,Caspase-8, NOX1 and LATS1 mRNA expression,and the lower the CyclinD1,Bcl-2,c-Myc,c-FLIP,TRAF6,N-cadherin and Vimentin mRNA expression.Conclusion:Cordycepin can promote pro-apoptosis gene expression and inhibit pro-proliferation and pro-in-vasion gene expression in lung cancer cell lines A549.
