各型FMDV VP1基因表位嵌入型蛋白SLP-MultiVP1对免疫不同小鼠血清IgG亚型的影响 被引量:1

Effect of various types FMDV VP1 gene epitope embedded protein SLP-MultiVP1 on IgG subtypes of different immune mice

摘要 利用通过Swiss-Model在线软件分析所得的乳杆菌S-层蛋白高级结构3D模拟图,应用Swiss-Pbd Viewer软件对嵌入型蛋白SLP-MultiVP1中的3型口蹄疫病毒(FMDV)不同表位位点进行标注,并用嵌入型蛋白SLP-MultiVP1和重组蛋白SLP分别免疫6周龄的BALB/c和昆明小鼠,分离3次免疫后的血清,用Mouse IgG1Ready-SET-Go和Mouse IgG2aReady-SET-Go ELISA试剂盒检测血清中IgG抗体亚型IgG1和IgG2a。通过SPSS统计学分析发现:嵌入型蛋白SLP-MultiVP1组IgG1和IgG2a水平显著高于SLP组(P<0.05),其中嵌入型蛋白SLP-MultiVP1免疫组的IgG2a显著高于IgG1(P<0.05),但是BALB/c和昆明小鼠之间无显著性差异(P>0.05)。该结果提示,嵌入型蛋白SLP-MultiVP1可能倾向于激发Th1类细胞介导的免疫应答,而且无个体差异。 In this study,the senior structure 3D rendering of Lactobacillus S-layer protein was analysed by online software of Swiss-Model,the three different FMDV epitope sites of embedded prorein SLP-MultiVP1 were marked by Swiss-Pbd Viewer software. Six-week-old BALB/c and Kunruing mice were vaccinated by embedded protein SLP MultiVP1 and recombinant protein SLP, PBS group was set as negative control. Third immunized serum of BALB/c and Kunming mice was detected for lgG1 and IgG2a of IgG antibody subtype by Mouse IgG1 Ready-SET-Go and Mouse IgG2a Ready-SET Go ELISA kit,SPSS statistical analysis found that the IgG2a and IgG1 of embedded protein SI.P-MuhiVP1 group were higher than that of SLP group(P〈0.05),IgG2a of embedded protein SLP MuhiVP1 group was higher than IgGI(P〈0. 05), there was no significant difference between BALB/c and Kunming groups(P〈0.05).
