转录组学分析鹅等级前卵泡中多胺代谢关键酶的差异表达 被引量:4

Transcriptome analysis of polaymine metabolic pathway in pre-hierarchical follicles of geese

摘要 为了解多胺代谢关键酶亚精胺合成酶(spermidine synthase,SRM)、精胺合成酶(spermine synthas,SMS)和精胺氧化酶(spermine oxidase,SMOX)在鹅等级前卵泡中的表达情况,本研究基于高通量测序技术对鹅等级前卵泡进行转录组测序分析,并通过荧光定量PCR对结果进行验证。通过FPKM值计算分析,这3个基因在鹅等级前卵泡中均有表达,且随着卵泡的发育,SRM和SMS的表达呈现先降低后增加再降低的趋势,在初级卵泡中的表达量达到最高值,在中白卵泡中的表达量最低;SMOX的表达则呈现先增加后降低的趋势,初级卵泡中的表达量最低,中白卵泡中的表达量最高。荧光定量PCR结果与转录组测序结果基本相同。结果证实SRM和SMS对卵泡的发育有促进作用,而SMOX可能参与卵泡闭锁过程。 Transcriptome sequencing technology was used to study the expression of the key en- zyme of polyamine metabolism,spermidine synthase(SRM),spermine synthas(SMS) and spermine oxidase (SMOX) in the pre-follieles of geese. The geese pre-hierarchieal follicles transcriptome was sequenced by using the Illumina Hiseq and fluorescence quantitative PCR. The result showed that the three genes all expressed in pre-hierarchical follicles,with the development of follicles,the expression of SRM and SMS displayed trend of firstly decrease and then increase and finally de creased, was the highest in the primary follicles and minimum in the middle white follicles. The expression of SMOX is first increased and then decreased,was the the highest expression in primary follicles and the minimum expression in the middle white follicles. The transcriptome result was consistent with fluorescent quantitative PCR,which confirmed that the SRM and SMS can promote the follicular development and the SMOX may involve in follicular atresia.
