芝麻油对大鼠脑缺血再灌注损伤的保护作用及其机制 被引量:3

Protective Effect of Sesame Oil Against Brain Ischemia Reperfusion Injury on Rats and Related Mechanism

摘要 目的:探讨芝麻油对大脑缺血再灌注损伤的保护作用及其作用的机制。方法:实验将雄性SD大鼠20只随机分为两组,对照组(C组,n=10),行大脑中动脉阻断前5 d及术前1 h予生理盐水4 m L/(kg·d)灌胃;芝麻油组(S组,n=10)于中动脉阻断前5 d及术前1 h给予芝麻油4 m L/(kg·d)灌胃。脑缺血90 min后恢复血流,于大脑中动脉阻断后24 h处死大鼠,测各组脑组织中核转录因子(Nuclear factor kappa B,NF-κB)P65表达阳性率(%)、丙二醛(malonaldehyde,MDA)和超氧化物歧化酶(superoxide dismutase,SOD)的水平;同时检测C组和S组的脑水肿指数、脑梗死的体积、神经功能评分。结果:脑梗死S组大鼠神经行为学评分明显低于C组,S组大鼠缺血侧的大脑水肿指数、脑梗死体积较C组明显改善(P<0.01):同时S组与C组比较,脑组织中核转录因子P65表达阳性率[(20.91±1.36)%vs(27.13±3.57)%]、MDA含量[(13.70±2.54)nmol/mg vs(3.96±0.68)nmo L/mg]均降低,SOD活性提高[(272.6±23.75)U/mg vs(189.5±16.53)U/mg],差异均有统计学意义(P<0.05)。结论:脑梗死期应用芝麻油可减轻脑组织的氧化应激反应,抑制炎症因子释放,从而对局灶性脑缺血后脑损伤具有保护作用。 Objective:To investigate the effect of sesame oil on the focal brain ischemia reperfusion injury and the related mechanism.Method:Twenty Sprague Dawley male rats were randomly divided into two groups:the control group was given middle cerebral artery occlusion(MCAO) and saline pretreatment for 5 days as placebo(n=10).Sesame oil group was given sesame oil via gastric gavage at a dose of 4 m L/(kg·d) for 7 days(n=10).Middle cerebral artery was occluded by thread for 90 min,followed by reperfusion.The neurological outcomes including edema index,infarc size and neurological deficit scores were evaluated 24 h after MCAO.The levels of malonaldehyde(MDA),the expression change of NF-κB/P65 and activity of superoxide dismutase(SOD) in brain tissue were assessed according to the corresponding kits.Result:Compared with C group,sesame oil significantly reduced the edema index,infarc size and neurological deficit scores at 24 h after MCAO(P〈0.01).Also,sesame oil decreased the tissue levels of the expression change of NF-κB/P65 [(20.91±1.36)% vs(28.45±3.57)% ],and MDA[(13.70±2.54)nmol/mg vs(3.96±0.68)nmol/mg,P〈0.05],and enhanced the activity of SOD[(272.6±23.75)U/mg vs(189.50±16.53)U/mg,P〈0.05] compared to C group.Conclusion:Sesame oil can protect against focal brainischemia reperfusion injury partly through the mechanism of attenuating oxidative stress and inhibiting the proinflammatory response.
