摘要
目的探讨脑源性神经营养因子(BDNF)及其低亲和力受体P75在ⅠB1~ⅡB期宫颈鳞癌及正常宫颈组织中的表达特点及表达位置关系。方法收集南方医科大学南方医院妇科2014—2016年ⅠB1~ⅡB期宫颈鳞癌病例124例,用免疫组织化学、免疫荧光及生物体视学图像分析法观察124例宫颈鳞癌及10例正常宫颈BDNF及P75的表达分布、定量情况及位置关系。结果 BDNF及P75受体均主要表达于宫颈鳞癌细胞及正常宫颈上皮细胞的细胞膜、其次为膜浆共表达。BDNF及P75的表达部位之间差异有统计学意义(χ2=4.127,P=0.042),两者表达位置之间无相关性(B=1.246,P>0.05)。在宫颈鳞癌中,BDNF的表达量低于正常宫颈(t=4.289,P=0.04);P75则高于正常宫颈(t=9.970,P=0.002)。BDNF、P75膜浆共表达组分期高于膜表达组(χ2=22.841,P=0.000)、(χ2=23.442,P=0.000)。各分期宫颈鳞癌BDNF及P75表达量之间差异无统计学意义(F=0.545,P=0.703)、(F=1.673,P=0.162)。结论脑源性神经营养因子及其低亲和力受体P75可能参与宫颈鳞癌的发生,且其两者之间存在一定关系,共同作用于肿瘤的生长。
Objective To study the characteristics of the expression of brain-derived neurotrophic factor and it's lowaffinity receptor P75 in early and middle stage cervical cancer and normal cervix,and to analyze its possible function in the development of cervical squamous carcinoma.Methods Immunohistochemistry,immunofluorescence and biological stereology were used to make a qualitative and quantitative analysis of the expression of BDNF and P75 in 124 cervical squamous carcinoma samples and another 10 normal cervix samples. Results BDNF and P75 mainly expressed in the cytomembrane and cytoplasm in cervical squamous carcinoma and normal cervix cells,followed by the co-expression of cytomembrane and cytoplasm;there was correlation existing between the expression site of BDNF and P75(χ2=4.127,P=0.042);the integral optical density(IOD)of BDNF expression in cervical squamous carcinoma was lower than that in normal cervix,while IOD of P75 was higher. For the co-expresson group,the general Figo stage was higher than the menbrane expression group(χ2=22.841,P=0.000)、(χ2=23.442,P=0.000).There existed no difference in expression quantity among different Figo stages,both for BDNF and P75,(F=0.545,P=0.703),(F=1.673,P=0.162).ConclusionBDNF and P75 may play a role in the development of cervical squamous carcinoma, while the two factors correlate with each other to a certain degree,which contributes to the progression of tumor.
