摘要
目的构建大鼠肺动脉高压右心衰竭模型,研究右心室Nogo的表达情况。方法将28只雄性SD大鼠随机分为对照组、肺动脉高压组、右心衰竭组。模型诱导采用野百合碱(MCT)60mg/kg一次性腹腔注射,生理盐水作为对照组。分别经右侧颈内静脉测量中心静脉压(CVP)。处死大鼠,称其肺脏(Lung Weight)、肝脏质量(Liver Weight,LW),计算出肺脏与体质量、肝脏与体质量的比值。去掉心房及大血管,游离右心室(RV)和左心室加室间隔(LV+S)后分别称重,并计算右心指数:RV/(LV+S)。同时观察右室心肌组织的Nogo的表达、Nogo蛋白合成和胶原生成情况。结果肺动脉高压组和右心衰竭组与对照组比较,肺动脉压力、中心静脉压、右室重量和右心指数进行性升高。肺动脉高压组和右心衰竭组NOGO-A mRNA逐渐升高,NOGO-B mRNA逐渐降低,CollagenⅠ和CollagenⅢmRNA随着右心室病变加重逐渐升高。Nogo-A蛋白表达量逐渐增加,Nogo-B1逐渐下降,Nogo-B2基本无变化。结论野百合碱注射法建立大鼠肺动脉高压右心衰竭模型可靠;Nogo在右心组织有表达,Nogo异常表达可能参与了右心衰竭的心室重塑。
Objective To construct the right heart failure model of pulmonary hypertension in rats, and to study the ex- pression of Nogo in the right ventricle. Methods 28 male SD rats were randomly divided into control group, pulmonary hyperten- sion group and right heart failure group. Model was induced by single intra-peritoneal injection of 60mg/kg monocrotaline( MCT), saline as substitution in control group. Central venous pressure (CVP)was measured through right internal jugular vein, respective- ly. The rats were executed and their lung weight was determinedas well as liver weight. Then ratios of lung weight to body weight and liver weight to body weight were calculated. As atria and great vessels were removed ,free right ventricle(RV) and left ventricle plus septum( LV + S) were weighed, respectively and right cardiac indexes were calculated as RV/( LV + S). Meanwhile, the ex- pression of Nogo, Nogo protein synthesis and collagen production were observed in the right ventricular myocardium. Results Compared with right heart failure group and control group, pulmonary hypertension group has progressively elevated pulmonary pressure, CVP, right ventricular weight and right cardiac index. In pulmonary hypertension group and right heart failure group, NO- GO-A mRNA increased gradually, while NOGO-B mRNA decreased gradually, and Collagen I and Collagen III mRNA increased gradually along with the exacerbation of right ventricular lesions. The expression of Nogo-A protein increased gradually, while No- go-B1 decreased and Nogo-B2 practically did not change. Conclusions It is credible to establish rat right heart failure model of pulmonary hypertension by injection of MCT. Nogo expresses in right heart tissues and the abnormal expression of Nogo may be in- volved in the ventricular remodeling of right heart failure.
