体外培育牛黄对小鼠慢性溃疡性结肠炎的疗效研究

Efficacy of Calculus Bovis Sativus on Chronic Ulcerative Colitis in Mice

目的:研究体外培育牛黄(CBS)对葡聚糖硫酸钠(DSS)诱导的慢性溃疡性结肠炎(UC)的治疗作用。方法:采用2%的DSS溶液连续给药3周期,在雄性C57BL/6小鼠中诱导慢性UC动物模型。造模的最后7 d连续给予50 mg·kg^(-1)和150mg·kg^(-1)的CBS灌胃给药处理,每日1次;通过小鼠一般情况、疾病活动(DAI)指数、结肠状态、HE染色和Tunel检测细胞凋亡等来评价CBS对DSS诱导慢性UC的疗效。结果:CBS减弱了DSS诱导的小鼠体质量降低、腹泻、便血等症状,并减少了其DAI指数,同时减轻了结肠的组织病理损伤。结论:CBS对于DSS诱导的慢性UC具有一定的治疗作用,值得进一步深入研究。

Objective： To study the efficacy of calculus bovis sativus （CBS） on chronic ulcerative colitis（UC） induced by dextran sulfate soduim （DSS） in mice. Methods： Male C57BL/6 mice were given 2% DSS solution for continuous three cycles to induce the chronic UC model. For the last 7 days of modeling, the mice were given CBS （50 and 150 mg kg -1 per day） intragastrically. The efficacy of CBS on chronic UC was evaluated by the general situation, disease activity index （DAI）, colon state, and HE staining and Tunel detection of cell apoptosis. Results： In the DSS-induced chronic UC mice, CBS alleviated the symptoms of weight loss, diarrhea, blood feces, and decreased the DAI index and reduced the pathological damage of colon. Conclusion： CBS has therapeutic effect on DSS-induced chronic UC in mice, which deserves further studies.
