摘要
目的观察CuSO_4对肝细胞癌(HCC)细胞株HepG2细胞核因子-κB(NF-κB)等相关炎症指标的影响。方法用含不同浓度CuSO_4的RPMI-1640培养液孵育HepG2细胞,四甲基偶氮唑盐(MTT)比色法检测其对细胞增殖的抑制作用;Western blot法检测不同浓度的CuSO_4对HepG2细胞炎症指标NF-κBp65、白细胞介素-1β(IL-1β)、白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、白细胞介素-6(IL-6)和γ干扰素(IFN-γ)等表达水平的影响。结果 CuSO_4在一定浓度范围内可抑制HepG2细胞的增殖;CuSO_4呈一定浓度依赖性地下调HepG2细胞中NF-κB、IFN-γ和IL-5蛋白的表达,上调IL-1β、IL-4和IL-6蛋白的表达。结论 CuSO_4能抑制HepG2细胞的增殖,并可通过下调NF-κBp65、IFN-γ和IL-5蛋白的表达并上调IL-1β、IL-4和IL-6蛋白的表达来抑制HepG2细胞的炎症反应。
Objective To observe the influences of copper sulfate on related inflammatory markers such as nuclear factor-KB (NF-KB) of HepG2 cells, the cell lines of hepatoeellular carcinoma (HCC). Methods HepG2 cells were cultured by RPMI-1640 contained various concentrations of copper sulfate in vitro. MTT colorimetric assay was used to detect the growth inhibitory effect of copper sulfate on HepG2 cell. Western blot was used to estimate the effect of copper sulfate on changes of related inflammatory markers, for instance, NF-KBp65, interleukin-1β (IL-1β) , interleukin -4 ( IL-4), interleukin-5 ( IL-5 ), interleukin-6 ( IL-6 ) and γ-interferon (IFN-γ). Results The growth of HepG2 cells was inhibited by copper sulfate in certain concentrations. The expression levels of NF-κB, IFN-γ and IL-5 were reduced and IL-1γ, IL-4 and IL-6 were up-regulated in HepG2 cells by copper sulfate with a dose- dependent manner. Conclusion Copper sulfate may inhibit the proliferation of HepG2 cells. And the inflammatory reaction in HepG2 cells can be suppressed by copper sulfate through lowering the levels of NF-κB, IFN-γ and IL- 5 and increasing the levels of IL-1β, IL-4 and IL-6.
